Applications

Our Abpromise guarantee covers the use of ab83135 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
Sandwich ELISA Use a concentration of 0.25 - 1 µg/ml.

Allows the detection of at least 0.2 - 0.4 ng/well of recombinant murine RANTES.

WB Use a concentration of 0.1 - 0.2 µg/ml. Predicted molecular weight: 10 kDa.

The detection limit for recombinant murine RANTES is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Target

  • Function

Chemoattractant for blood monocytes, memory T-helper cells and eosinophils. Causes the release of histamine from basophils and activates eosinophils. Binds to CCR1, CCR3, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant RANTES protein induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form RANTES(3-68) acts as a natural chemotaxis inhibitor and is a more potent inhibitor of HIV-1-infection. The second processed form RANTES(4-68) exhibits reduced chemotactic and HIV-suppressive activity compared with RANTES(1-68) and RANTES(3-68) and is generated by an unidentified enzyme associated with monocytes and neutrophils.
  • Tissue specificity

    T-cell and macrophage specific.
  • Sequence similarities

Belongs to the intercrine beta (chemokine CC) family.
  • Post-translational
    modifications

N-terminal processed form RANTES(3-68) is produced by proteolytic cleavage, probably by DPP4, after secretion from peripheral blood leukocytes and cultured sarcoma cells.
The identity of the O-linked saccharides at Ser-27 and Ser-28 are not reported in PubMed:1380064. They are assigned by similarity.
  • Cellular localization

    Secreted.

      Western blot - Biotin Anti-RANTES antibody (ab83135)

To detect murine RANTES by Western Blot analysis ab83135 can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant murine RANTES is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Image shown is reducing conditions; Murine recombinant RANTES protein at 250 - 0.24 ng/ml.

Lane 1 is MW marker, lanes 2-12 murine recombinant RANTES protein.

      Sandwich ELISA - Biotin Anti-RANTES antibody (ab83135)

To detect mouse RANTES by sandwich ELISA (using 100 μl/well antibody solution) a concentration of 0.25 – 1.0 μg/ml of ab83135 is required. This biotinylated polyclonal antibody, in conjunction with a appropriate anti-RANTES antibody as a capture antibody, allows the detection of at least 0.2 – 0.4 ng/well of recombinant mouse RANTES.

    Question

    Dear Sir/Madam,

I hope that you are fine and well on receiving this message. I would like to order a antibodies for detection of bovine CCL5 (RANTES) and GCP-2 (CXCL6) using ELISA. We would like to develop our own ELISA detection and therefore would like to know what primary (capture) antibody and secondary (detection) antibody could be used for this purpose. I have looked at your website and found some antibodies but unsure whether it would detect bovine sequences. Some of the antibodies stated that they detected either human, mouse or rat.

If you could suggest appropriate antibodies for the described purpose, then it would be much appreciated.

I look forward to your reply.

    Best regards,

    Answer

    Thank you for contacting us.

The protein sequence compatibility between human and mouse with bovine is about 80% which is very close for antibody cross reactivity. We unfortunately haven't tested any of the antibody with bovine CCL5 so I am sorry we are unable to confirm their suitability. However as the homology is higher and therefore possibility of cross reactivity so the anti human might react with Bovine also. We can suggest following antibodies as pair

ab83135 as capture - ab9915 as detection antibody.
ab83324 as capture antibody - ab71978 as detection (this ab need to be conjugated)

CXCL6; the possible combinations could be

    ab109783 (capture) and ab83138 (detection)

Please note that none of these antibodies are actually tested with the bovine samples. Our assumption about the Bovine cross reactivity is totally based on the 5age sequence homology between human and bovine.

As these antibodies are not tested with Bovine sample so these are eligible for 1

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
