Target

  • Relevance

    S.aureus binds IgG molecules by their Fc region. In serum, the bacteria will bind IgG molecules in the wrong orientation on their surface, which hinders opsonization and phagocytosis. Mutants of S. aureus lacking protein A are more efficiently phagocytosed in vitro, and mutants in infection models have diminished virulence. Due to its affinity for the Fc region of many mammalian immunoglobulins, protein A is considered a universal reagent in biochemistry and immunology.

    Thank you for your response and follow up inquiry. Protein A on the surface will interact non-specifically with this antibody, as well as other Ig isotypes which can interact with Protein A. If you would like to minimize this interaction, you can block their sample with normal serum prior to primary antibody incubation. Normal serum from the species of the secondary antibody would be best. 10% normal serum incubated for about 10-15 minutes should help.

    Thank you for your inquiry regarding 0300-0085. For this specific antibody, the method of purification is proprietary but I can say that it is greater than 95% purity. The antibody has been tested against both soluble and structural antigens of the whole bacterium, so it is expected to react with Protein A.

    Just to clarify, you have 1 vial labeled ab35192 and the other labeled ab35192-1? You had listed ab35912 which is a Histone H1.4 peptide no longer available on the catalogue, so I think this was just a typo.

    The -1 just signifies the size (1 mL). This was an older method of labeling that we used to employ, but newer labels have dropped the size from the catalogue number so you won't see the -1 anymore (it was confusing to customers). But yes, they are the same product.

    Our customer has some problem with abcam's antibodies.
    Her application is similar ELISA but, not guaranteed one so, she needs to check ELISA test to prove antibodies' activity.

    The anti-Staphylococcus aureus antibodies ab68954, ab35192 and ab20002 have not all been tested in ELISA and cannot therefore be guaranteed in this application. Ab35192 has currently only been used in ICC/IF but this does not mean that it would not work in this application, simply that we have not tested it and do not know if it will be suitable.

    With the Anti-Staphylococcus aureus antibody [702] (ab20002), the antibody was used in a direct ELISA protocol using the heat killed Salmonella organisms directly to the plate. The cells were not lysed prior to coating on the plate. The Staph. aureus cells (5 x 107/ml) were bound to the wells of Immulon II 96-well plates in 50ul of 100 mM carbonate buffer, pH 9.6, per well.The plates are sealed with plastic wrap and incubated overnight at room temperature. The next day, the coating solution is removed by inverting the plates. The wellswere then blocked for 30 minutes with 1% BSA-PBS-0.05% Tween 20. This solution was then removed by inverting the plates.The coated plates are then ready for testing with the antibody.

    Please find below the information I managed to collect by contacting the sources of the Anti-Staphylococcus aureusand the Anti-Streptococcus agalactiaeantibodies we have in our catalog :

    Anti-Staphylococcus aureus:
    https://www.abcam.com/ab23568 shows no reactivity to any streptococcus sp., mycoplasma sp. or corynebacteria sp.

    Unfortunately, ab35192 has not been specifically tested against streptococcus so we are unable to guarantee it won’t react to S. agalactiae.

    ab68950, ab68952, ab68954: These Staphylococcus aureus polyclonal antibodies are not absorbed for related microorganisms and so therefore could cross react with related bacteria such as Staph. Epidermidis.

    MLS : Staphylococcus aureus: We do not have an antibody that is specific to Staph. aureus but does not have crossreactivity to Staph. epidermidis.

    Anti-Streptococcus agalactiae:
    https://www.abcam.com/ab21126 has weak crossreactivity with Strep. dysgalactiae and does not cross-react with Strep. uberis, Strep. bovis, Strep. faecalis, Staph. aureus, Mycoplasma bovis, Mycoplasma californicum, Mycoplasma bovigenitalium, E. coli or Corynebacterium bovis.
