FunctionComponent of the Polycomb group (PcG) multiprotein PRC1 complex, a complex required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility. In the PRC1 complex, it is required to stimulate the E3 ubiquitin-protein ligase activity of RNF2/RING2.

Lanes 1 - 4: Merged signal (red and green). Green - ab155069 observed at 42 kDa. Red - loading control, ab176560, observed at 52 kDa.
ab155069 was shown to specifically recognize Bmi1. Wild-type and Bmi1 knockout samples were subjected to SDS-PAGE. ab155069 and ab176560 (loading control to alpha tubulin) were diluted at 2.5 ug/mL and 1/10 000 respectively and incubated overnight at 4°C. Blots were developed with Goat anti-Mouse IgG H&L (IRDye® 800CW) preadsorbed ab216772 and Goat Anti-Rabbit IgG H&L (IRDye® 680RD) preadsorbed ab216777 secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.
All lanes : Anti-Bmi1 antibody [LLBmi1-1] (ab155069) at 1 µg/ml (blocked with 3% milk)
Lane 1 : MOLT4 (Human acute lymphoblastic leukemia cell line) Whole Cell Lysate
