This monoclonal antibody to Bmi1 has been knockout validated in Western blot. The expected band for Bmi1 was observed in wild type cells and the band was not seen in knockout cells.

FunctionComponent of the Polycomb group (PcG) multiprotein PRC1 complex, a complex required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility. In the PRC1 complex, it is required to stimulate the E3 ubiquitin-protein ligase activity of RNF2/RING2.

Lanes 1 - 4: Merged signal (red and green). Green - ab155069 observed at 42 kDa. Red - loading control, ab176560, observed at 52 kDa.
ab155069 was shown to specifically recognize Bmi1. Wild-type and Bmi1 knockout samples were subjected to SDS-PAGE. ab155069 and ab176560 (loading control to alpha tubulin) were diluted at 2.5 ug/mL and 1/10 000 respectively and incubated overnight at 4°C. Blots were developed with Goat anti-Mouse IgG H&L (IRDye® 800CW) preadsorbed ab216772 and Goat Anti-Rabbit IgG H&L (IRDye® 680RD) preadsorbed ab216777 secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.
This data was developed using the same antibody clone in a different formulation containing PBS and Azide (ab155069).
