Application Abreviews Notes
WB 1/1000 - 1/5000. Predicted molecular weight: 111 kDa. A recommended starting concentration for Western blots is 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 for chemiluminescent substrates. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen.

  • Function
Cleaves the C-terminal propeptides of procollagen I, II and III. Induces cartilage and bone formation. May participate in dorsoventral patterning during early development by cleaving chordin (CHRD). Responsible for the proteolytic activation of lysyl oxidase LOX.
  • Tissue specificity
  • Involvement in disease
    Osteogenesis imperfecta 13
  • Sequence similarities
Belongs to the peptidase M12A family.
    Contains 5 CUB domains.
    Contains 2 EGF-like domains.
  • Post-translational
Proteolytically activated in the trans-Golgi network by furin-like/paired basic proprotein convertases, cleavage is not required for secretion.
  • Cellular localization
Golgi apparatus, trans-Golgi network. Secreted, extracellular space, extracellular matrix. Co-localizes with POSTN in the Golgi.

Thank you very much for your interest in ab38953. To our knowledge, ab38953 has not been tested in rat. Unfortunately I can not provide a sequence alignment neither, as the immunogen sequence is proprietary information of the laboratory producing this antibody and I do not have access. I am very sorry. I have however aligned the whole sequence of mouse to the rat sequence (as we know this antibody reacts with mouse) and the sequences seem to be very conserved. http://www.uniprot.org/align/2011082560LBQDGUB8 So I would suggest that crossreactivity is likely. However, this needs to be assessed experimentally. If you would like to test this, I can offer a discount off a future purchase if you buy ab38953 now, test it in rat and submit feedback to us in the form of an Abreview. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback. The discount would be to the value of 1 free primary antibody. If you are interested in this offer, please follow these steps: 1. Reply to this e-mail to let me know that you would like to proceed and test ab38953 in rat. I will then send a discount code. This code must be issued before purchasing ab38953 so please wait for my reply before ordering. 2. Purchase ab38953 either by phone, fax, or online (www.abcam.com). 3. Test it in rat. 4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out more about our Abreview system, please visit: https://www.abcam.com/abreviews. 5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any primary antibody ordered and the discount code is valid for 4 months after issue. We are always pleased to obtain feedback about our products and any information is greatly appreciated! Even if ab38953 turns out to be unsuitable for rat, you will still receive the discount on your next purchase after your Abreview has been submitted. Please let me know if you have any questions about this offer and I would be happy to help you further. The Terms and Conditions of this offer can be found at: www.abcam.com/collaborationdiscount.
