Application Abreviews Notes
WB Use a concentration of 1 - 2 µg/ml. Detects a band of approximately 17-18 kDa.
ELISA Use at an assay dependent concentration.
IHC-P Use at an assay dependent concentration. Do not perform antigen retrieval. PubMed: 17959825
ICC/IF Use a concentration of 1 µg/ml.
Indirect ELISA Use a concentration of 0.5 - 1 µg/ml. To detect a limit of ~3 ng/well recombinant human BMP-2 at 50 and 100 ng/lane under non-reducing and reducing conditions, respectively.



  • ICC/IF image of ab6285 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab6285, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of rabbit periosteum labeling BMP2 with ab6285 at 1/100 dilution. The tissue was fixed with paraformaldehyde, followed by blocking in 5% serum for 20 minutes. The tissue was incubated with ab6285 for 12 hours at 4°C in antibody dilution solution. An undiluted polyclonal goat-anti mouse HRP conjugate secondary antibody was used.

  • ab66285 staining BMP2 in Human small intestine tissue sections by IHC-P (formaldehyde-fixed paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 1.5% serum for 10 minutes. Antigen retrieval was by heat mediation in citrate (pH6.0). Samples were incubated with primary antibody (1/100 in diluent buffer) for 1 hour. An undiluted HRP-conjugated Goat polyclonal to mouse IgG was used as secondary antibody

These conditions will vary among our antibodies, therefore, it is important to verify the storage conditions for each of our products when you receive them. We guarantee all of our products to work for at least 180 days from the date of purchase when stored correctly.

I can confirm that both ab6285 and ab69498 should be aliquoted and stored at -20C or below. It will not make a difference if they are stored at -20C or -80C,but it is important to avoid freeze/thaw cycles.

I have had the opportunity to speak with the lab to try and clarify the statement,"The antibody recognizes recombinant human BMP-2. It shows approximately 25% cross-reactivity with recombinant human BMP-4."They feel this should be interpreted as that the BMP-4 signal is a quarter of the strength of the BMP-2 signal.

BMP-2 is abundant in lung, spleen and colon and so any of these tissues should, in theory, be suitable positive controls. To the best of my knowledge this antibody was not tested with native human tissue. The antibody was characterized using recombinant human BMP-2 and recombinant human BMP-4, so multiple banding is not likely to have been observed during QC analysis. Unfortunately we do not have any images available for this particular product.

With regards to ab6285 Anti-BMP2 antibody [65529.111], I have reviewed the alingment of the immunogen for this antibody to the Zebrafish protein. There is 63% alignment to zebrafish. This alignment is quite low and I would therefore not recommend that this antibody is likely to detect the Zebrafish BMP2. The other antibodies we have to BMP2 are raised against full length human protein or shorter peptides from the human sequence, and they all have the same alignment or less. I am sorry there are no BMP2 antibodies to suggest that may be suitable on this occasion. With regards to BMPR2, the immunogen of ab78422 BMPR2 has only 58% alignment to zebrafish. Again, I am sorry this means it is not likely to crossreact with Zebrafish. However, we do have the following BMPR2 antibody which has 88% alignment to the zebrafish protein.
