      Brain natriuretic peptide (BNP) circulates in blood as a peptide hormone with natriuretic, vasodilatory and renin inhibitory properties. BNP is secreted predominantly by the left ventricular myocytes in response to volume expansion and pressure overload. BNP belongs to a family of structurally similar peptide hormones, which includes atrial natriuretic peptide (ANP), BNP, C type natriuretic peptide (CNP) and urodilatin. These peptides are characterized by a common 17 amino acid ring structure with a disulfide bond between two cystein residues. This ring structure shows high homology between different natriuretic peptides (eleven of the 17 amino acid residues are homologous in the ring of each of the natriuretic peptides). BNP is a 32 amino acid peptide with disulfide bond between the cysteine residues Cys10 and Cys26. In earlier studies it has been demonstrated that BNP concentration in blood increases with the severity of congestive heart failure. Quantitative measurement of BNP in blood provides an objective indicator of congestive heart failure severity.

    Dear Sir/Madam,

    I am working as a Research Associate at the Mawson Institute (University of South Australia).We are working onthe detection ofbrain natriuretic peptide (BNP) and troponin I.Regarding BNP, currently we are working with a glycosylated proBNP (108aa) with the following amino acid sequence:

    We already have an antibody (from mouse) specific to the region 26-32 (GKLSELQ), that we are using as the detection antibody. Now we are looking for another antibody specific to either the region 11-17 (DLETSGL) or 11-22 (DLETSGLQEQRN) that should be from another source (not mouse, since it is going to be the capture antibody of an indirect sandwich ELISA).

    I have been told by one of our colleagues that you sell a monoclonal rabbit anti-BNP that could be used for our purpose. The reference is ab92500 and it has been raised against an immunogen derived from within amino acids 75 - 110 of human BNP. Although this antibody is not specific for one of the regions that I mentioned before, I would be very grateful if you could confirm us that this antibody will be suitable as capture antibody in a sandwich assay where the recognition antibody is specific to the region GKLSELQ.

    Additionally, we would like to buy some antibodies against human cardiac troponin I. We are interested in antibodies specific to either the region 18-28 or 83-93 of human cardiac troponin I, and also from a host different to mouse.

    I would appreciate if you can confirm the suitability of any of your antibodies for our work and, in such case, send us a quotation for their purchase.

    As shown on the attached file (ab92500 v proBNP.pdf), ab92500seems to be suitable for your work as it would bind to the proBNP at a different region than the Mouse antibody that binds the region aa 26-32.
