Target

    Clostridium botulinum neurotoxins are zinc-dependent proteases that block the release of acetylcholine from peripheral cholinergic nerve endings by targeting specific membrane proteins including synaptosomal-associated proteins (SNAP), vesicle-associated membrane proteins (VAMP), and syntaxins. Neurotoxin A is characteristically associated with human botulism and known to target and hydrolyse the 197-Gln-|-Arg-198 bond of SNAP-25.

Answer


The antibody was raised against formaldehyde inactivated Type A Hemagglutinin 33, thus I am not certain if the sequence has been determined for the immunogen. The epitope has definitely not been mapped.

I will check with the lab if any additional information is available and let you know.

I just heard back from the supplying laboratory that yes, the immunogen for ab20436, Rabbit anti C. botulinum Type A Hemagglutinin-33, was purifed prior to injecting the rabbit. I have a few questions regarding the products ab20436 & ab20641. Firstly, what is the difference between these two antibodies? I see that one of them says specificity for Hemagglutinin, but don't realize the difference. I would like to use this to do IHC on BOTOX TYPE A. Is it possible to use one of these antibodies for this? Also, could you kindly email me the detection limits (either in ng/ml or U/ml). Thank you so much for your help. I would really appreciate a prompt response.

Answer

Thank you for your enquiry and for your interest in our products. Clostridium botulinum neurotoxin is produced along with a group of neurotoxin-associated proteins which protect the neurotoxin from low pH and proteases of the gastrointestinal tract. The neurotoxin together with the neurotoxin-associated proteins make up the neurotoxin complex. Hemagglutinin-33 is one of the neurotoxin-associated proteins. Ab20436 is specific for C. botulinum Hemagglutinin-33. Ab20641 is specific for the C. botulinum Neurotoxin. These antibodies have not been tested in IHC. I do not have any information about BOTOX cosmetic and the detection limits have not been determined using these antibodies.
