Binds to target promoters, leading to increased histone H3 acetylation at 'Lys-9' (H3K9ac). Binds to the ESR1 promoter. Recruits BRCA1 and POU2F1 to the ESR1 promoter. Coactivator for TP53-mediated activation of transcription of a set of target genes. Required for TP53-mediated cell-cycle arrest in response to oncogene activation. Promotes acetylation of TP53 at 'Lys-382', and thereby promotes efficient recruitment of TP53 to target promoters. Inhibits cell cycle progression from G1 to S phase.

    why is the actual band size different from the predicted?

  • ICC/IF image of ab56036 stained MCF7 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab56036, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

These two antibodies (ab46553, ab56036) you mentioned are polyclonal antibodies. Polyclonal antibodies consist of a mixed population of IgGs each of which will recognize a different epitope. Generally, epitope mapping experiments for polyclonal antibodies are not performed - only for monoclonal antibodies.

Regarding the immunogen sequence, if it is not commercially sensitive information, we can publish it on-line on the product datasheets. Unfortunately, we are unable to release proprietary information and this is the case for ab56036.

ab46553 and ab56036 are from two different sources and their manufacture process, purification and characterization (tested applications) are different as well. The prices of our antibodies reflect all of these factors.
