  • Relevance

    Brevican is the most abundant chondroitin sulfate proteoglycan in the extracellular matrix of the adult brain. It is a member of the lectican family of aggregating extracellular matrix proteoglycans that bear chondroitin sulfate (CS) chains. It is highly expressed in the central nervous system and is thought to stabilize synapses and inhibit neural plasticity. Brevican is secreted from astrocytes and neurons as a 145 kD core protein that bears up to three, covalently-linked, CS chains. It is also is secreted as a 145 kD core protein without CS chains. When cleaved by extracellular glutamyl endopeptidases, the ADAMTSs, a 55 kD N-terminal fragment is formed that contains the unique C-terminal epitope EAMESE.

Since you obtained poor results using the antibody in an untested application, we would like to follow up on this to see if we can possibly improve the results you are seeing with this antibody.

Based on the information in your review, I feel that altering the following step in your protocol may help improve your results:
- try another fixation method (e.g. methanol or acetone)
- do a no primary control to check how much background comes from the secondary antibody alone
- I can't tell for sure if a confocal microscope was used for your images. If not, it might be worthwhile trying, since you would be able to move the focal plane through the cell and could be able to see the extracellular staining better and distinguish between cytoplasmic background and membrane/ECM staining.

Do you happen to know which region of Brevican is membrane bound?The immunogen for ab111719 falls within the region betweenamino acids 83 and 262 of human Brevican. According to the SwissProt entry for this protein (Q96GW7), this region encompasses a portion of the Ig-like V-type domain, the entire link 1 domain and a portion of the link 2 domain. However, I am unsure if any of these domains are involved in membrane binding.
