    BrightMount/Plus is an aqueous-based, ready to use mounting medium for cover slipping tissues stained with fluorescent dyes. This product is compatible with FITC, phycoerythrin, phycocyanin and allophycocyanin. It can be also be used with other fluorescent markers such as Texas Red and rhodamine. The anti-fading agent preserves strong staining, allowing the user to view slides several months after they have been stained.

    Mounting Procedure

    • Remove the red tip from the bottle and cut the tip off with a sharp razor or scissors. This will help acheive smooth flow of BrightMount/Plus and prevent formation of tiny bubbles.
    • Place the bottle upside down in a container before using. This will help clear BrightMount/Plus of tiny bubbles.
    • Blot excess water from the slide without letting tissue dry. Make sure the tissue is wet prior to mounting.
    • Apply 2-3 drops of BrightMount/Plus on the tissue section and gently apply a cover slip. Allow to dry for 30-45 minutes at room temperature. Seal edges of the cover slip with nail polish for long term storage.

    Removal of BrightMount/Plus
    BrightMount/Plus is an aqueous-based mounting medium and can easily be removed by soaking slides in deionized water. Place slide in a beaker full oe deionized water on a magnetic stirrer. Leave the slide stirring in water for a few hours or overnight for complete removal of dry BrightMount/Plus.

For the experiment the customer is planning to perform, the BrightMount (ab103746) or BrightMount/Plus (ab103748) would be most appropriate and would be compatible with the fluorescent dyes the customer wants to use. The BrightMount/Plus provides the added advantage of containing an anti-fading agent which preserves strong fluorescent staining, allowing the user to view slides months after they have been stained.

The supplying lab of our EasyLink kits doesn't carry out microscopy, so they haven't testing this themselves.Theyresearched AMCA’s degree of stability against photobleaching/fading, and confusingly different sources say different things: that it is resistant to photobleaching, that it has moderate photobleaching, or that it fades rapidly; that it retains its fluorescence much longer than fluorescein, or that fades as rapidly as fluorescein. Many sources do, however, recommend that an anti-fading agent is used with AMCA. Thereare a variety of anti-fading agents available, so you could say that the exact rate of fading will depend on the particular conditions selected.

Having spoken with one of my colleagues I can now confirm that the reagent if ready prepared to be added to the tissue slide. All you need to do is make sure that the tissue is covered by the medium, so around 100-200 uL should be sufficient.

We currently do not have any product against the C.elegans protein MKS-5 nor will our products against a similar proteinRPGRIP1L recognize MKS-5. A protein with a similar name is MSK5X (also known as CD99). We do have a large selection of antibodies against this target. Ab8855 (https://www.abcam.com/CD99-antibody-12E7-ab8855.html) has been tested in ICC/IF while ab108297 (https://www.abcam.com/CD99-antibody-EPR3096-ab108297.html) and ab75858 (https://www.abcam.com/CD99-antibody-EPR3097Y-ab75858.html) are both guaranteed to work in ICH and ICC on human samples.

Differences in the chemical compositions of the mounting medium make using one product for both IHC and IF difficult. The conditions required for excellent perioxidase preservation are not neccessarily good for the anti- photobleaching requirements of IF. As such I cannot recommend a single medium for you customer but suggest they by mounting mediums specific to their needs.

Our standard mointing medium (ab64230;https://www.abcam.com/Mounting-Medium-ab64230.html) is appropriate for IHC.



For IF I recommend BrightMount/Plus (ab103748;https://www.abcam.com/BrightMount-Plus-25ml-Aqueous-Mounting-Medium-Anti-fading-for-Fluorescent-Staining-ab103748.html)
