    Abcam’s BSA Removal Kit (ab173231) is a simple one-step, 10 minute method which effectively separates the BSA from the antibody. The antibody is left in a suitable position for transfer to a buffer more suited to conjugation. The BSA Removal Kit can be used on any antibody sub-type, and species.

    This 1 ml kit can remove all of the BSA from up to 1.25 ml of antibody, with a BSA concentration of 0.5% or less. For higher BSA  concentrations, the method may need to be repeated, or a higher volume of BSA Removal Buffer may be required.

    Please note that this kit is not compatible with our range of GOLD conjugation kits. To remove BSA from antibodies prior to conjugation with our GOLD conjugation kits please use GOLD BSA removal kit (ab204912).

    Bovine Serum Albumin (BSA) is often added to purified antibodies as it is an effective stabilizer. However, when labelling antibodies, the BSA becomes a hindrance, as it directly competes with the antibody to attach to the label, greatly reducing the conjugation efficiency. Therefore, prior to undertaking labelling techniques, it is essential to remove the BSA. Common commercial BSA removal techniques can involve many laborious steps.

The ab173231 BSA Removal Kit purification system is based on the molecule size. We have never tested the kit for an already-conjugated antibody.

Was the antibody conjugated to FITC using our Abcam FITC conjugation kit? If so, the way it processes the molecules of FITC might affect the solubility of the protein in this salt-induced precipitation. We have never tried putting any pre-conjugated protein through the kit so we cannot really guarantee a successful purification with the BSA removal kit.

If the antibody was already conjugated to FITC using a different kit working with different technology, I cannot say for sure if it will work. The only main consideration here is how the molecule with interfere with salt-based precipitation.

I’m sorry I can’t be of much more help, If you do decide to try the kit I will be interested to hear from you with an update on how successful it was.

The BSA removal kit would not be appropriate for in this case because cell culture medium will contain many other proteins which will not be removed by this kit

To purify the antibody from tissue culture supernatant I can suggest to consider one of the purification kits.

I am sorry I do not have details of the particular antibody you are purifying, but the following sound be suitable for purification from tissue culture supernatant. I can recommend to review the details to see if it would be suitable and let me know kf you have any questions.

The lab have confirmed that the BSA removal kit ab173231 will be suitable for removing Albumin from both the rat and monkey serum samples. It is however worth noting that, due to the nature of the purification, it is probable that some of the other buffer components will also be removed.
