Target

  • Function
Displays several functions associated with host defense: it promotes agglutination, bacterial capsular swelling, phagocytosis and complement fixation through its calcium-dependent binding to phosphorylcholine. Can interact with DNA and histones and may scavenge nuclear material released from damaged circulating cells.
  • Tissue specificity
    Found in plasma.
  • Sequence similarities
    Belongs to the pentaxin family.
    Contains 1 pentaxin domain.
  • Cellular localization
    Secreted.
  • Information by UniProt

Images

  • Calibration curve using ab8277 [C2] (biotinylated) and ab8278 [C6] (labelled with stable Eu3+ chelate) in a sandwich ELISA. Mixture of antibodies and antigen (10 µl sample) were incubated for 10 minutes at R.T. in streptavidin coated plates.

  • Sensitivity to EDTA. Serum samples with (dark blue) and w/o (light blue) Ca2+ ions were tested by two two-site combinations of anti-CRP MAbs. Ab10028 [C4] is sensitive to the conformational changes of CRP molecule and thus the assay gives no signal in the presence of EDTA. Sandwich immunoassay utilizing ab8277 [C2] and ab8278 [C6] recognizes the antigen with the same efficiency in the presence and in the absence of EDTA.

Answer

Thank you for contacting us.

I am sorry we do not have any data to confirm how many ab molecules binds to CRP. Theoretically, I agree there is possibility of binding 5 ab molecule per pentamer of CRP however we can't be sure as we do not know for how many chains the epitope is exposed for antibody binding.

I am sorry due to non availability of tested we will not be able to confirm this.

I hope this information is nevertheless helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Answer

Thank you for contacting us. Dr. Tanya is out of office today so I am dealing with her inquiries.

The immunogen for these antibodies is a proprietary information. These antibodies are raised sing full length human CRP protein. The two products are tested positive in sandwich ELISA.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Thank you very much for your reply.

It will be no problem to ship it after August 20. We are actually expecting to have this antibody in stock by August 27, so it should arrive shortly after that date. The antibody will be arriving free of charge on the order ***.

So that we can look into the trouble with these antibodies, could you also briefly describe the protocol used with ab8278 and ab8279 in sandwich ELISA?

I look forward to hearing from you. Thank you very much and have a nice day!

Answer

Thank you again for letting us know about the trouble with the CRP antibodies in ELISA.

We now have the unconjugated C2 clone in our catalog as ab136176, and I'd be happy to send a free of charge vial for your to try.

Could you please let me know your original order or PO number and I will send this out promptly?

Thank you for your patience in resolving this issue, and please let me know if there is anything else that we can do for you.

Answer

Thank you very much for your reply.

I agree that the differences in the protocol may be contributing to the background you're experiencing. I'm working with my colleagues to see if we can provide the unconjugated C2, and I will update you as soon as I have more information about this antibody.

We do not have a full protocol for testing blood plasma CRP protein, and since the titer of this protein is variable, it would be a good idea to try a few dilutions of your plasma samples to determine the optimal dilution. I would recommend using at least a 1:1 dilution to reduce any possible matrix effects that can obscure the specific signal.

The DELFIA washing buffer is only necessary for the specific one-step immunoassay that the lab ran with the Eu-conjugated detection antibody, and I don't think that this buffer will improve the results in a more standard ELISA. PBS or PBST should be a sufficient washing buffer to reduce non-specific binding.

I will be back in touch with you once I've heard from my colleagues about the C2 clone. Please let me know if you have any questions in the meantime. Thank you for your patience!

Answer

Thank you very much for your call earlier this week and for letting us know about the trouble with these antibodies.

I've heard back from the lab, and these antibodies were actually tested together in a one-step fluoroimmunoassay;please see the full protocol attached. This protocol is not a standard sandwich ELISA protocol, and I'm not sure that it will be helpful as a referencefor the kind of assay that you are running.

Based on the information that you've given me, I'd be happy to send a different antibody to try in the assay. We only have the HRP-conjugated C2 clone in our catalog at this time, but I cancheck whetherweare able tosend a vial of unconjugated C2 if you'd like. If there is a different CRP antibody that you would like to try, I can send that instead.

I look forward to hearing from you. Please let me know if there is an antibody or pair of antibodies that you would like to try, or if you have any questions about this information.

Answer

Thank you for contacting us.
We do not use the terminology C number. These does not depicts any information other the clone numbers for a particular antibody.
The sELISA pairs we recommend are
The best pair is C2-C6 (ab24461 detection - ab8278 capture)
C5-C6 (ab8279 capture - ab24462 detection)
C7-C6 (ab8280 capture - ab24462 detection)
Please ab24462 is a HRP conjugated anti C reactive protein antibody; due to the conjugation it will be a good detection antibody.
I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Thank you for contacting Abcam. If you wish to place a bulk order I can happily forward your request to in house specialist how deals with these matters. However we generally only do custom batches for larger orders. If you just want to increase the concentration of the antibody, then I would recommend using one of our concentration kits: https://www.abcam.com/Antibody-Concentration-Kit-3-columns-ab102778.html Please let me know if you would like me to pass your information onto our bulk specialist. If there is anything else I can help you with do not hesit

Thank you for your enquiry. For the curve on our datasheet, the following amounts of antibody were used: C2-biotinylated: 100 ng/well (100 ul) C6-Eu (Europium) labeled: 200 ng/well (50 ul) Please do note that the amounts and concentrations depend on the application, so this information should be considered only as starting point suggestions for the optimization.
