  • General notes

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Applications

  • Application notes
    Is unsuitable for IP.
  • Target

    • Function

Receptor for the chemotactic and inflammatory peptide anaphylatoxin C5a. This receptor stimulates chemotaxis, granule enzyme release and superoxide anion production.
    • Sequence similarities

Belongs to the G-protein coupled receptor 1 family.
    • Post-translational
      modifications

Sulfation plays a critical role in the association of the receptor with C5a, but no significant role in the ability of the receptor to transduce a signal and mobilize calcium in response to a small peptide agonist.
    • Cellular localization

      Cell membrane.

      Question

Inquiry: I notice that in the latest batch of ab11884 the storage recommendations have changed from 4 degrees to 'aliquot and store at -20 degrees'. Is this due to some sort of change in the formulation or recent testing indicating better preservation of the antibody at this temperature? Thanks

      Asked on Jun 18 2012

      Answer

      Thank you for contacting us.

The formulation for this product has not changed. Recent tests showed thatab11884 can be stored at 4ºC as well as at -20ºC, this is why the storage information has been updated. In general the life timeof an antibody storedat -20ºC is about 5 years. As shown on the datasheet, there is no need to dilute the product but it is recommended to aliquot it and to avoid freeze / thaw cycles.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

      Answer

I have spoken to one of our scientists in the lab who advised me that to perform blocking experiments we always use the immunogen without its conjugation, therefore just the synthesized peptide should be suitable for your purpose. I was advised that we have had good results in peptide synthesis from a company called CSS, I hope this information will be useful. I also noticed that we did have access to the immunogen and have added the sequence clearly on the datasheet. I would like to also suggest you submit an Abreview with your results. We always encourage customers to send their results back to us, whether positive or negative, and we make all product information available to researchers. We reward each Abreview with Abpoints which you can exchange for Amazon gift vouchers, conference attendance and many gifts. To find out more about our Abreview system, please see the following URL: https://www.abcam.com/abreviews I wish you all the best with your research, please feel free to call me if you need further assistance,

      Question

I would like to know which region of C5aR does the antibody is raised against? As I'm looking for an anti-C5aR (CD88) antibody that doesn't not targets ligand binding site, i.e. intracellular region.

      Answer

The antibody was raised against the first extracellular domain of the protein termed "topo_dom" on the SwissProt database which you can access via our datasheet. I hope this information helps, please let me know if you need more details,
