    Constituent of the membrane attack complex (MAC) that plays a key role in the innate and adaptive immune response by forming pores in the plasma membrane of target cells. C9 is the pore-forming subunit of the MAC.

    why is the actual band size different from the predicted?

    We hypothesize that the 70 kDa band represents the glycosylated C9 protein (predicted molecular weight 63). Under appropriate conditions this antibody should also recognise the C9a cleavage product.

Both antibodieswere raised against an immunogen from C9 (a component of the C5b-9 complex).

We recommend ab55811 for specifically detecting the C5b-9 complex.

After discussing this matter with my colleagues in our lab, we have decided to change the target for this antibody from C5b-9 to C9. The antibody was raised against a sequence from the C9 component of the C5b-9 complex. While it is possible that this antibody may recognize the complex as well as C9, our testing does not currently prove that explicitly. The WB band we observe corresponds to C9 because it was tested under reduced and denatured conditions. Under native conditions, it is possible that ab71330 would detect the complex at a much higher molecular weight, but this has not been tested in house.
My colleagues are currently expanding our testing for this antibody in ICC and IHC and hope to provide more information about whether or not it detects the complex in the near future. I hope this helps, please let me know if you need any additional information or assistance.
