ab11426 staining Calbindin in human brain tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and antigen retrieval was by heat mediation in a EDTA buffer. Samples were incubated with primary antibody (1/5000) for 20 minutes hours at 25°C. A Goat anti-mouse anti-rabbit IgG was used as the secondary antibody.

ab11426 staining Calbindin in Mouse embryonic brain and kidney tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 and blocked with 1% BSA for 1 hour at 20°C. Samples were incubated with primary antibody (1/1000) for 1 hour at 20°C. An Alexa Fluor488®-conjugated Goat anti-rabbit IgG polyclonal (1/1000) was used as the secondary antibody. Specifically detects Purkinje cells in developing cerebellum and distal renal tubules in kidney.

NB: No labeling observed following omission of primary antibody.
Sections were viewed using an Axioplan 2 Imaging microscope (Imaging Associates) fitted with 10x, 20x and 40x Plan-Neofluorobjectives (Zeiss, Germany) and images were taken using a AxioCam Hrm digital camera (Zeiss, Germany) and AxioVision software (Imaging Associates).
