We have only two antibodies anti-human calcitonin receptor guaranteed to work in WB in our catalogue: ab103422 and ab11042.

Both of these should recognise all three human isoforms since there is a large sequence homology between all isoforms. However, we have not directly tested this and therefore cannot guarantee this.

I have just received the following information from the source of ab11042: "The cross-reactivity of this antibody with the human calcitonin receptor has been demonstrated by the antibody originator using human foetal kidney (pathology specimens), fixed in formalin. The antibody was successfully used at dilutions as low as 1:500 but a catalysed signal amplification (CSA) system was required (also called tyramide signal amplification). Antigen retrieval was not required, indeed the originator did not have much success when employing such techniques. Based on this information, we would suggest that the customer employs this kind of signal amplification step on her renal biopsies. The originator reports staining of osteoclasts (rodent) using the polyclonal antibody ab11042 both in paraffin sections and from cell culture. To our knowledge, human osteoclasts have not been specifically tested. This antibody has recently been tested on COS7 cells transfected with rat calcitonin receptor and human calcitonin receptor. Not surprisingly, this anti-rat antibody reacts best with the rat calcitonin receptor, and less so with human calcitonin receptor (antibody reactivity and working conditions may vary between species). These studies were conducted on fixed permeabilised COS7 cells, using immunofluorescence. We would recommend the use of the mouse anti-human calcitonin receptor antibody ab11040. Again this antibody has been used successfully on human foetal kidney sections (formalin-fixed, paraffin) with signal amplification may improve the staining. Perhaps your customer would consider testing this antibody in their system, as this would be more suitable for their needs." I hope this information will be of some assistance to you.

I'm sorry to hear you are having problem with ab11042. I would like to suggest trying the immunohistochemistry steps without antigen retrieval prior to staining of paraffin embedded sections. I would also recommend using triton (0.3%v/v)in PBS in all buffers used to block/dilute antibodies/dilute streptavidin.
