Ich kann empfehlen den ab87597 mit einer Verdünnungreihe, ausgehend von 1/100 )zum Beispiel 1/50, 1/100 und 1/200) auszutested. Auch kann ich empfehlen, die Antigendemaskierung mit einem Citrate Puffer pH 6 unter Hitze vorzunehmen. In der Tat gab dieser Antikörper gute Resultate wenn er benutzt wurder mit 3.8 µg/ml auf human Hirncortex Gewebe. Wir empfehlen jedoch immer, den Antikörper in Ihren Konditionen und auf dem von Ihnen benutzten Gewebe auch zu optimisieren.
Der ab105502 wurder erfolgreich benutzt, um humanes Brustkrebsgewebe einzufärben. Dies wurde mit einem Leica BondTM System gemacht (Protocol F). Da wurde das Gewebe vorbehandelt mit einer Demaskierung mit Citrate pH 6 unter Hitze für 20 Minuten. Der ab105502 wurde dann mit einer Verdünnung von (5µg/ml) benutzt. Wie auch für den anderen Antikörper, kann ich empfehlen, diese Angaben als Ausgang zu benutzen und die Verdünnung und Demaskierugn in Ihrem Labor zu optimisieren - speziell auch die Länge der Erhitzung im Demaskierungspuffer hängt stark vom Gewebe und dessen Fixierung ab.
Der ab22131 wurde nicht in paraffin Schnitten getestet, sondern auf einfachen PFA fixierten Schnitten. Das Protokoll war etwas speziell - Sie können die Details auf dem Datenblatt unter dem Bild nachlesen. Um diesen Antikörper auf den Paraffinschnitten zu testen, kann ich Ihnen empfehlen eine Verdünnungsreihe von 1/50, 1/100 und1/200 empfehlen, wie auch Hitze Antigendemaskierung mit Citrate pH6 oder EDTA pH 9 auszuprobieren.
