Target

  • Function

    Calcium/calmodulin-dependent protein kinase belonging to a proposed calcium-triggered signaling cascade involved in a number of cellular processes. Isoform 1, isoform 2 and isoform 3 phosphorylate CAMK1 and CAMK4. Isoform 3 phosphorylates CAMK1D. Isoform 4, isoform 5 and isoform 6 lacking part of the calmodulin-binding domain are inactive. Seems to be involved in hippocampal activation of CREB1.
  • Tissue specificity

    Ubiquitously expressed with higher levels in the brain. Intermediate levels are detected in spleen, prostate, thyroid and leukocytes. The lowest level is in lung.

    Belongs to the protein kinase superfamily. Ser/Thr protein kinase family.
    Contains 1 protein kinase domain.
  • Domain

    The autoinhibitory domain overlaps with the calmodulin binding region and may be involved in intrasteric autoinhibition.
    The RP domain (arginine/proline-rich) is involved in the recognition of CAMKI and CAMK4 as substrates.

    I have a customer enquiry about antibodies to CAMKK and LKB1, application = WB / IF, species = dictyostelium

    I know that you do not have antibodies to these targets tested in Dictyostelium, but I had trouble finding out about identity of the proteins between the target species and Dicty. So I am hoping you can get some information in a more expedient fashion that I can pass onto the customer.

    I guess it might be possible that at Trial (testing) discount might be applicable?

    Thank you for contacting us.

    I have performed a sequence alignment for LKB1 for Dictyostelium and for human (which is used to raise most of our antibodies.) Unfortunately there is only a total of 33% homology between the two proteins. I have contacted the lab and indeed all the immunogens of the anti-LKB1antibodies guaranteed to work in WB/IF in our catalogue have a 28-50% sequence homology with the Dictyostelium protein and therefore are not likely to work.

    The amino acid sequence for LKB1 for Dictyostelium that I used can be found at http://www.uniprot.org/uniprot/Q15831.fasta. Unfortunately, this is the predicted amino acid sequence for the species and has not been confirmed therefore, if your customer has the verified sequence of this protein, then I will be happy to repeat these alignments to verify the homology.

    Regarding an anti-CAMKK antibody, I am not able to recommend a suitable one unless I have the Dictyostelium amino acid sequence for this protein. If your customer could send it, then again I will be happy to perform the sequence alignments.

    I have found the amino acid sequence for LKB1 for Dictyostelium which can be found at http://www.uniprot.org/uniprot/Q15831.fasta

    I have contacted the lab to see if any of our antibodies would be compatible for use in Dictyostelium.

    I have not been able to find the sequence for CAMKK for Dictyostelium. Could you ask your customer to send it if they have it so that I may perform the sequence alignments?
