Both antibodies should recognize the pro form and the fragment downstream of the K158/T159 cleavage site. ab62484 detects a band about 45kDa in human heart and ab62463 detects a band about 40kDa in mouse and rat liver.

I am happy to confirm that all these antibodies are tested and guaranteed to work with human samples.
You are correct that we do not have any reports yet about the use of these antibodies in muscle. I assume that is your customers area of research. It is not possible for us to test our product with all available tissues and therefore I would like to re-assure you and your customer that these antibodies are covered by our Abpromise guarantee when positive controls beside muscle are used in experiments.
I would recommend doing this anyway especially if your customer does not know what to expect in muscle as it is part of a research project.

Usinga single antibody, the only way to distinguish between the pro and cleaved forms of Caspase 12 would be by the molecular weight in a western blot. The full length pro Caspase 12 will run at ˜42 kDa, while the long cleavage product will have a molecular weight of ˜38 kDa.

In IHC, you would need to perform a co-stain with an antibody specific for the other cleavage fragment. Caspase 12 is cleaved at residue 120 into the long and small cleavage products. By using ab62484 (raised against the long cleavage product) and ab62463 (raised against the small cleavage product), you will be able to determine the localization of several forms of Caspase 12. Areas stained by only ab62484 will show only the long cleavage product andareas stained by only ab62463 will show only the small cleavage product. Areas showing co-localization will either correspond to the full length pro Caspase 12 or to cleaved Caspase 12, where the long and small forms are co-localized. Unfortunately, it is very difficult to detect the pro form specifically in IHC without an antibody directed against the cleavage site, which we do not currently sell.

You may also be able to tell the difference between the pro and cleaved forms of Caspase 12 if you have an idea of where the cleavage products translocate to within the cell. I hope this helps, please let me know if you need any additional information or assistance.
