Experiments were performed on Rhesus macaque vaginal tissue biopsies under various parameters, including tissue that had been freshly frozen in OCT tissue that had originally been fixed in Formalin and then frozen and tissue that had been fixed and frozen, then treated with 1% Triton-X for 1 minute for permeabilization. Overall, the fixed (frozen) sections showed the best response, and these parameters are detailed in the review. Unfortunately, though we were expecting the labeling of vaginal smooth muscle cells, instead we saw staining of fibroblasts and the vascular endothelium only. The image included shows nuclei counterstained with DAPI (in blue). Examples of muscle bundles are circled in white, which contain multiple individual myocytes that should have been labeled.
