Target

  • Function
Receptor for cholecystokinin. Mediates pancreatic growth and enzyme secretion, smooth muscle contraction of the gall bladder and stomach. Has a 1000-fold higher affinity for CCK rather than for gastrin. It modulates feeding and dopamine-induced behavior in the central and peripheral nervous system. This receptor mediates its action by association with G proteins that activate a phosphatidylinositol-calcium second messenger system.
  • Sequence similarities
Belongs to the G-protein coupled receptor 1 family.
  • Cellular localization
    Cell membrane.

Images

  • All lanes : Anti-CCK1-R antibody (ab28627) at 1/1000 dilution

    Lane 1 : Mouse Pancreas
    Lane 2 : Rat Pancreas

Lysates/proteins at 30 µg per lane.

    Secondary
All lanes : Goat anti-Rabbit IgG (H+L) HRP conjugate at 1/2500 dilution

Observed band size: 47 kDa (why is the actual band size different from the predicted?)
Additional bands at: 80 kDa (possible isoform)

  • Immunofluorescence analysis of CCK1-R was performed using 70% confluent log phase PANC-1 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with CCK1-R Rabbit Polyclonal Antibody ab28627 at 1/250 dilution in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor® 488 conjugate at a dilution of 1/2000 for 45 minutes at room temperature (a: green). Nuclei (b: blue) were stained with DAPI. F-actin (c: red) was stained with Rhodamine Phalloidin (1/300). Panel d represents the merged image showing membranous localization. Panel e shows the no primary antibody control.
