Application Abreviews Notes
WB Use a concentration of 1 - 2 µg/ml. Detects a band of approximately 40 kDa (predicted molecular weight: 38 kDa).
ICC/IF Use a concentration of 5 µg/ml.
IHC-P Use a concentration of 5 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

Images

  • ICC/IF image of ab47682 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab47682, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

  • IHC image of ab47682 staining in human normal breast formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab47682, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

The exact sequence is proprietary, but the immunogen overlaps the amino terminus of the region specified, starting a little before it.

My colleague from datasheet department has confirmed that they never changed the target name for this product it was always CCNO. So could you confirm how you have received this antibody when you ordered anti-UNG2 antibody? I also have checked the Fax we received from Nottingham University which clearly mention anti CCNO antibody cat#ab47682 so it seems the error happened when the university ordered this antibody. Unfortunately we charge 20% restocking fees when products mistakenly ordered by and the return delivery charges have to bear by customer.

You being a valued customer I am happy to replace the antibody and waive off the charges. Could you send the product back to us by overnight delivery.

When you return the goods, please ensure that you follow the procedures as below:

- Return in the original Abcam box with the cooler box and ice pack (or similar) within 7 days from today.

- Return all documents (or copies of) that arrived with the product (if they’re available – should be invoice, delivery note and datasheet). After conducting further research about he target I have to say the antibody is specific to CCNO. Swissprot ID P22674.

The UNG2 and CCNO target were used as similar in past. However this antibody is agianst CCNO not UNG2. I apologize for the confusion.
