Receptor for a C-C type chemokine. Binds to eotaxin, eotaxin-3, MCP-3, MCP-4, RANTES and MIP-1 delta. Subsequently transduces a signal by increasing the intracellular calcium ions level. Alternative coreceptor with CD4 for HIV-1 infection.
  • Tissue specificity
In eosinophils as well as trace amounts in neutrophils and monocytes.
  • Sequence similarities
Belongs to the G-protein coupled receptor 1 family.
  • Cellular localization
    Cell membrane.

Images

  • ICC/IF image of ab16231 stained Hek293 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab16231, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Answer

ab16231's immunogen is human CCR3 transfected into mouse B cell lymphoma L1.2 cells. I have performed a sequence alignment between the bovine and human sequences and found a 72% total homology suggesting that there may be cross-reactions with this antibody. Looking through the literature, I found the following paper which I think may be of interest to you: http://www.biomedcentral.com/1471-2164/11/439

This group confirmed the coding sequences for CCR-3. Below are the links to the predicted bovine CCR3 sequences which I used to perform the alignments:
http://www.uniprot.org/uniprot/D9ZDE0
http://www.ncbi.nlm.nih.gov/protein/303227943?report=fasta

By comparing the human and bovine sequence, the only part of the protein which is dissimilar is the N terminus so only an antibody who's immunogen is against the N terminus could work.

Unfortunately in our catalogue none of our antibodies guaranteed to work in human in IHC-Fr are directed towards the N terminus of the protein. We do have ab1667 which is directed to the N terminus and who's immunogen shares a 40% homology with the human sequence but this is not guaranteed to work in IHC-Fr so this product would not be covered by our guarantee. If you would be willing to test these on human cells only (not human cells in bovine tissue) we could give you a testing discount and you could test human cells alone before testing human cells in bovine tissue.

With regards to CXCR1, I have compared the bovine and human sequences and again found a 72% total sequence homology with the only dissimilar region again being the N terminus. See publication http://www.ncbi.nlm.nih.gov/pubmed/22056270 which compares the sequences.

ab60254 is guaranteed to work in human and in IHC-Fr and its immunogen is between amino acids 5-19 which shares about 20% homology with human so is unlikely to cross- react with bovine however we cannot guarantee this.

Answer

Thank you very much for your interest in our anti-CCR3 antibodies. To our knowledge, none of ouranti-CCR3 antibodieshas not been tested in Flow Cytometry as well as in IHC-P. Therefore, I can offer a discount off a future purchase if you buy an anti-CCR3 now, test it in IHC-P or Flow Cytometryand submit feedback to us in the form of an Abreview. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback. The discount would be to the value of 1 free primary antibody. You couldbuy for example, ab16231 (https://www.abcam.com/ab16231)and test it in IHC-P, or an anti-CCR3 tested in IHC-P and test it in Flow Cytometry. The link to the list of thefour anti-CCR3 tested in IHC-P and humanis : https://www.abcam.com/index.html?pageconfig=searchresults&search=CCR3&pt=1&sk=app&sv=74&sn=IHC-P&l=3&fViewMore=1. If you are interested in this offer, please follow these steps: 1. Reply to this e-mail to let me know that you would like to proceed and test ab***** in IHC-P or Flow Cytometry. I will then send a discount code. This code must be issued before purchasingthe antibody to be testedso please wait for my reply before ordering. 2. Purchase antibody to be tested either by phone, fax, or online (www.abcam.com). 3. Test it in IHC-P or Flow Cytometry. 4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: https://www.abcam.com/abreviews. 5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any primary antibodyordered and the discount code is valid for 4 months after issue. We are always pleased to obtain feedback about our products and any information is greatly appreciated! Even ifthe tested antibodyturns out to be unsuitable for IHC-P or Flow Cytometry, you will still receive the discount on your next purchase after your Abreview has been submitted. Please let me know if you have any questions about this offer and I would be happy to help you further. The Terms and Conditions of this offer can be found at: https://www.abcam.com/collaborationdiscount. Regarding the anti-CCR10, I confirm that ab3904 has been testedand is guaranteed to work in Flow Cytometry, IHC-P and on Human samples.
