    Receptor for a C-C type chemokine. Binds to eotaxin, eotaxin-3, MCP-3, MCP-4, RANTES and MIP-1 delta. Subsequently transduces a signal by increasing the intracellular calcium ions level. Alternative coreceptor with CD4 for HIV-1 infection.

  • ICC/IF image of ab16231 stained Hek293 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab16231, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

ab16231's immunogen is human CCR3 transfected into mouse B cell lymphoma L1.2 cells. I have performed a sequence alignment between the bovine and human sequences and found a 72% total homology suggesting that there may be cross-reactions with this antibody. Looking through the literature, I found the following paper which I think may be of interest to you: http://www.biomedcentral.com/1471-2164/11/439

This group confirmed the coding sequences for CCR-3. Below are the links to the predicted bovine CCR3 sequences which I used to perform the alignments:

By comparing the human and bovine sequence, the only part of the protein which is dissimilar is the N terminus so only an antibody who's immunogen is against the N terminus could work.

Unfortunately in our catalogue none of our antibodies guaranteed to work in human in IHC-Fr are directed towards the N terminus of the protein. We do have ab1667 which is directed to the N terminus and who's immunogen shares a 40% homology with the human sequence but this is not guaranteed to work in IHC-Fr so this product would not be covered by our guarantee. If you would be willing to test these on human cells only (not human cells in bovine tissue) we could give you a testing discount and you could test human cells alone before testing human cells in bovine tissue.

With regards to CXCR1, I have compared the bovine and human sequences and again found a 72% total sequence homology with the only dissimilar region again being the N terminus. See publication http://www.ncbi.nlm.nih.gov/pubmed/22056270 which compares the sequences.

ab60254 is guaranteed to work in human and in IHC-Fr and its immunogen is between amino acids 5-19 which shares about 20% homology with human so is unlikely to cross- react with bovine however we cannot guarantee this.
