    Inquiry: I am having trouble getting signal using your CCR2 (ab32144, lot GR52022-3), CCR3 (ab32512, lot GR52022-3), and CCR5 (ab32048, lot GR40445-5) antibodies. Here are my Western Blotting conditions: 1. Loaded 75, 50, 30, 20, and 10 micrograms per well of mouse spleen extract under both reducing and non-reducing conditions on a 4-12% Bis-tris gel from Invitrogen. MES buffer was used for the run. 2. Proteins were transferred onto a nitrocellulose membrane using an iBlot system (Invitrogen). 3. Membrane was blocked for 1 hour RT in 5% milk 4. Primary incubation for all three antibodies was overnight (approx. 16 hours) at 4 C. Primary antibodies were diluted 1:500 in 5% milk. 5. Secondary incubation was using goat anti-rabbit from santa cruz (1:5,000) for 1 hour RT. 6. Membrane was washed following both primary and secondary incubations three times in PBS-T. 7. West Pico was used to develop the blot, and bands were visualized using an ImageQuant LAS4000, and exposures were for 30 minutes. Our primary goal is to quantify changes in CCR2, 3, and 5 in brain and brain endothelial cells, however, to optimize antibody conditions, we typically use some other tissue type as a positive control. For CCRs, spleen seemed suitable. We see very a very faint band for CCR2 which appears to be at the correct molecular weight, but IR is very weak. We did not detect any bands for CCR5, and the bands observed for CCR3 were not the correct molecular weight. That all three antibodies did not work as expected is of concern to me. I'd appreciate your assistance with this issue.

    Thank you also for sending so many details of your protocol.The protocol seems to be ok and similar to the protocol that the lab uses to test this antibody, and I would expect to see bands at the correct molecular weights under these conditions. Does the anti-rabbit secondary antibody work with other primary antibodies? Was protein transfer confirmed with a Ponceau stain or some other method? If you probed for a loading control like actin, what were the results?

    In general we recommend to use EDTA pH9. This antigen retrieval method is optimal for the most antigens. Please see the attachment and this publication (Emoto et al., J Histochem Cytochem. 2005 Nov ;53 (11) : 1311-21.) for more information.

    ab28775 was used successfully by our customers without, with citrate buffer pH6 and with Tris pH10. Please see Abreviews on the datasheet.

    The image on the datasheetfor ab32512 was generated with citrate buffer pH6.

    The optimal protocol usually has to be determined by the end user individually.
