Binding was detected using the secondary antibody, Alkaline Phosphatase-conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) at 1/2500 dilution.

Immunohistochemical analysis of paraffin-embedded human tonsil tissue labeling CCR6 with ab227036 at 1/1000 dilution, followed by the ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Membranous staining on human tonsil (PMID: 12514792) is observed. Counterstained with hematoxylin.
Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is the ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101).

CCR6 was immunoprecipitated from 0.35 mg HL-60 (Human acute promyelocytic leukemia promyeloblast) whole cell lysate with ab227036 at 1/30 dilution. Western blot was performed on the immunoprecipitate using ab227036 at 1/500 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366) was used for detection at 1/1000 dilution.
Lane 1: HL-60 whole cell lysate 10 µg (input).
Lane 2: ab227036 IP in HL-60 whole cell lysate.
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab227036 in HL-60 whole cell lysate.Blocking and dilution buffer and concentration: 5% NFDM/TBST.
Exposure time: 1 second. -
