Target

  • Function

    Receptor for CCL19 and chemerin/RARRES2. Does not appear to be a signaling receptor, but may have a role in modulating chemokine-triggered immune responses by capturing and internalizing CCL19 or by presenting RARRES2 ligand to CMKLR1, a functional signaling receptors. Plays a critical role for the development of Th2 responses.
  • Tissue specificity

    Expressed abundantly in immunal tissues such as spleen, fetal liver, lymph node and bone marrow. Strong expression also in lung and heart. Expressed in almost all hematopoietic cells including monocytes, macrophages, PMNs, T-cells (both CD4+ and CD8+), monocyte-derived iDCs, NK cells, and CD34+ progenitor cells. B cells expressed isoform 1 but not isoform 2. Up-regulated on synovial neutrophils of rheumatoid arthritis patients.

Since you obtained poor results using the antibody in a tested species and application, we would like to follow up on this to see if we can possibly improve the results you are seeing with this antibody.
Based on the information in your review, I feel that altering the following step in your protocol may help improve your results:
- use positive control that expresses the protein in high copy number. The cells that you have gated might not express a lot of the protein under the conditions studied and thus, only a small shift is seen.
- incubate for a longer time with the primary antibody
If you decide to follow these suggestions, please let me know if they are helpful. Additionally, this antibody is covered by our Abpromise, so if the suggestions do not help and the antibody was purchased within the past 6 months, I would be happy to discuss a refund or replacement with you to resolve this issue.

Since you obtained poor results using the antibody in a untested species (mouse), we would like to follow up on this with some protocol tips:
- try a different isotype control (I am surprised that this isotype control you used gives some specific signal at around 10^3)
- incubate longer with the priamry antibody
