  • Function

    Major glycoprotein of vascular endothelium. May play a critical role in the binding of endothelial cells to integrins and/or other RGD receptors.

    Endoglin is restricted to endothelial cells in all tissues except bone marrow.

    Defects in ENG are the cause of hereditary hemorrhagic telangiectasia type 1 (HHT1) [MIM:187300, 108010]; also known as Osler-Rendu-Weber syndrome 1 (ORW1). HHT1 is an autosomal dominant multisystemic vascular dysplasia, characterized by recurrent epistaxis, muco-cutaneous telangiectases, gastro-intestinal hemorrhage, and pulmonary (PAVM), cerebral (CAVM) and hepatic arteriovenous malformations; all secondary manifestations of the underlying vascular dysplasia. Although the first symptom of HHT1 in children is generally nose bleed, there is an important clinical heterogeneity.

    Cells were fixed with formaldehyde, permeabilized with Triton X and blocked with 3% BSA for 1 hour at 25°C. Samples were incubated with primary antibody (1/200) for 1 hour at 25°C. An undiluted FITC-conjugated Goat anti-rabbit IgG polyclonal was used as the secondary antibody.

The details provided by you will enable us to investigate this case and will provide us with vital information for monitoring product quality. I appreciate the time you have spent in the laboratory and understand your concerns. It is regrettable the results have not been successful. I have looked into exactly how the anitbody has been used in Western blotting and the protocol is very similar to that which you have already been using (please refer to the attachment of this email). I therefore do not think that it is this which is causing the problems encountered. I believe this product should have given satisfactory results. It appears that you may have received a faulty vial.

Since you obtained poor results using the antibody in a tested species and application, we would like to follow up on this to see if we can possibly improve the results you are seeing with this antibody.

Based on the information in your review, I feel that altering the following step in your protocol may help improve your results:

1) try a diferent antigen retrieval step (e.g. Tris/EDTA or enzymatic)
2) incubate longer with the primary antibody (e.g. overnight at 4 degree)
3) use more primary antibody (e.g. 5 ug/ml)
4) check that the secondary antibody is working well otherwise

If you decide to follow these suggestions, please let me know if they are helpful.
