Additional Notes
IHC/Fr image of ab52478 stained THP-1 cells (red). Four cell types (EA.hy926 below A549, THP-1 and HMC-1 above) were cultured on membrane inserts, frozen in Tissue-Tek® and cut into 14 µm sections. Sections were fixed for 5 min in 4% formaldehyde at ambient temperature (RT). Blocking of unspecific reactions was performed with 5% donkey serum in PBS for 30 min at RT. THP-1 were then stained with ab52478 (1:60) for 1 h at RT and secondary anti-rabbit-Cy3 (1:250, red) for another hour at RT. Cytoplasm of EA.hy926 and A549 were counterstained with Evans Blue (blue). Nuclei were labelled with Yopro (green) and the section was mounted with ProLong® Diamond Antifade. The picture shows an enlarged section of the membrane with THP-1 labelled in red.
