Immunofluorescent analysis of 80% methanol fixed and 0.1% TritonX-100 permeabilized HDLM-2 (human Hodgkin lymphoma) labeling CD137 with ab300349 at 1/500 dilution followed by preadsorbed Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488, Green, ab150081) at 1/1000 dilution. Confocal image showing cytoplasmic and membranous staining in HDLM-2 cell line.
Negative control: Jurkat (PMID: 25409517).Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594, Red, ab195889) at 1/200 dilution was used as a counterstain. Nuclear counterstain is DAPI.

Flow cytometric analysis of Jurkat (Human T cell leukemia T lymphocyte, Left) / HDLM-2 (Human Hodgkin lymphoma, Right) cells labeling CD137 with ab300349 at 1/500 dilution (Red) compared with isotype control Rabbit monoclonal IgG (ab172730) / Black and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue).
Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/2000 dilution was used as secondary antibody (Green).

CD137 was immunoprecipitated from HDLM-2 (human Hodgkin lymphoma) whole cell lysate 10 μg with ab300349 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab300349 at 1/1000 dilution. VeriBlot for IP secondary antibody (HRP) (ab131366) at 1/5000 dilution was used as a secondary antibody.
Lane 1 (Input): HDLM-2 (human Hodgkin lymphoma) whole cell lysate 10 μg
Lane 2 (+): ab300349 IP in HDLM-2 whole cell lysate
Lane 3 (-): Rabbit monoclonal IgG (ab172730) instead of ab300349 in HDLM-2 whole cell lysate
Exposure time: 15 seconds.
