    ab17051 is very useful for macrophage phenotyping. Positive staining can be seen in the skin (histiocytes), gut, Kupffer cells, few aveolar macrophages, main population of macrophages in the placenta, varying degrees of macrophages in inflammed tissue including tumorous tissue depending on the inflammatory stage. Red pulp, not white pulp, macrophages in the spleen and cortical macrophages of the thymus are detected. Antibody is sensitive to ionic detergents and to chloroform/methanol treatment.

      Acute phase-regulated receptor involved in clearance and endocytosis of hemoglobin/haptoglobin complexes by macrophages and may thereby protect tissues from free hemoglobin-mediated oxidative damage. May play a role in the uptake and recycling of iron, via endocytosis of hemoglobin/haptoglobin and subsequent breakdown of heme. Binds hemoglobin/haptoglobin complexes in a calcium-dependent and pH-dependent manner. Exhibits a higher affinity for complexes of hemoglobin and multimeric haptoglobin of HP*1F phenotype than for complexes of hemoglobin and dimeric haptoglobin of HP*1S phenotype. Induces a cascade of intracellular signals that involves tyrosine kinase-dependent calcium mobilization, inositol triphosphate production and secretion of IL6 and CSF1. Isoform 3 exhibits the higher capacity for ligand endocytosis and the more pronounced surface expression when expressed in cells.
      After shedding, the soluble form (sCD163) may play an anti-inflammatory role, and may be a valuable diagnostic parameter for monitoring macrophage activation in inflammatory conditions.

      Expressed in monocytes and mature macrophages such as Kupffer cells in the liver, red pulp macrophages in the spleen, cortical macrophages in the thymus, resident bone marrow macrophages and meningeal macrophages of the central nervous system. Expressed also in blood. Isoform 1 is the lowest abundant in the blood. Isoform 2 is the lowest abundant in the liver and the spleen. Isoform 3 is the predominant isoform detected in the blood.

      The SRCR domain 3 mediates calcium-sensitive interaction with hemoglobin/haptoglobin complexes.
    • Post-translational
      modifications
      A soluble form (sCD163) is produced by proteolytic shedding which can be induced by lipopolysaccharide, phorbol ester and Fc region of immunoglobulin gamma. This cleavage is dependent on protein kinase C and tyrosine kinases and can be blocked by protease inhibitors. The shedding is inhibited by the tissue inhibitor of metalloproteinase TIMP3, and thus probably induced by membrane-bound metalloproteinases ADAMs.

    Generally, each antibody and batch should be titrated to determine the optimal working dilutions/concentrations.

    ab17051: this antibody has only been tested in ICC/IF and IHC/Fr but it is unsuitable for formalin-fixed paraffin-embedded sections. The unit size is 50 µg at 0.1mg/ml and it is suggested to apply for IHC-Fr at aconcentration of 4 µg/ml (please read application notes).

    ab21562:this antibody has only been tested in IHC/Fr but it is unsuitable for formalin-fixed paraffin-embedded sections. The unit size is 100 µg at 0.5mg/ml but the working dilution needs to be optimized by the end user.

The other antibodies for CD163 have not been tested with rat so far. I do not have the rat sequence to check sequence homology. If you could sent me the sequence, I'd be happy to check the homology between human and rat for you. And if the homology is 85% or higher (giving a good chance of cross reactivity), you can try one of the antibodies with rat samples and use our testing discount program.     Thank you for your enquiry. I am sorry to the delay to your enquiry and sorry to hear that you have been having difficulties with this antibody. I have been in touch with the originator and they have offered the following suggestions. CD163 is a membrane glycoprotein, which is expressed exclusively on the cell surface of human monocytes and macrophages. Therefore no antigen retrieval step should be necessary. CD163 is sensitive to ionic detergents and chloroform/methanol treatment, this is not just for Western Blot but also for IHC-P protocols. Can I ask whether you tested another antigen on the cells as positive control, to determine whether your procedure was ok? At this stage I would like to suggest that you perform the test one more time without antigen retrieval using an overnight incubation.
