    Defects in MS4A1 are the cause of immunodeficiency common variable type 5 (CVID5) [MIM:613495]; also called antibody deficiency due to CD20 defect. CVID5 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B cells is usually in the normal range, but can be low.

The cells incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab1169, 0.06 µg/1x106 cells) for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG2a FITC (1 µg/1x106 cells ) for 30 min at 22°C. Acquisition of >5,000 events was performed.

Thank you for your enquiry. Although ab27856 has not been epitope mapped, the originator believes that it recognizes a region on the extracellular surface of CD20 because no permeabilization treatment is required prior to staining. For ab1169, the immunogen was cells of lymphoblastoid cell line DAUDI. It is strictly specific to an extracellular epitope of human CD20 antigen, present on normal peripheral B lymphocytes, malignant cells of pre-B acute lymphoblastic leukaemia patients and malignant cells of B-type chronic lymphoblastic leukaemia patients. It is not necessary to permeabilize the cells. I have added this information to the datasheets.
