  • General notes

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Question

I thought it would be easier just to ask for help instead of ordering the wrong thing. We're doing a T cell isolation from mouse spleens and need anti-CD3 and anti-CD28 for the culture plates. My boss asked me to get prices for the 500ug size.

Answer

Thank you for contacting us.

While we do not have any antibodies that are specifically tested for your purposes, I would recommend ab113628 for CD3. This antibody recognizes an extracellular epitope and gives good results in ELISA and Flow Cytometry, which are similar to your assay. For CD28, we currently have two clones that recognize extracellular epitopes and are reactive with mouse: PV-1 (ab25234) and 37.51 (ab25233). These clones are available directly conjugated to a variety of fluorophores and biotin.

If you will be purchasing more than five vials in total, my colleagues in customer services can help you determine if you are eligible for a discount when you place your order. If you will be purchasing more than 1mg of a single antibody, our bulk sales team can help prepare a quote for you: mailto:sales@abcam.com

I hope this helps, please let me know if you need any additional information or assistance.

Answer

Thank you for contacting us. Clone PV-1 is approved for In vitro costimulation of mouse T cells and NK cells, though we do recommend using the LE/AF format when working with live cells. Product #ab83051 has been tested to ensure low endotoxin levels and is supplied in PBS, pH 7.4. I have added the lnk to ab83051 below: https://www.abcam.com/CD80-antibody-AP-M-Low-endotoxin-Azide-free-ab83051.html I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
