    Defects in CD3D are a cause of severe combined immunodeficiency autosomal recessive T-cell-negative/B-cell-positive/NK-cell-positive (T(-)/B(+)/NK(+) SCID) [MIM:608971]. A form of severe combined immunodeficiency (SCID), a genetically and clinically heterogeneous group of rare congenital disorders characterized by impairment of both humoral and cell-mediated immunity, leukopenia, and low or absent antibody levels. Patients present in infancy recurrent, persistent infections by opportunistic organisms. The common characteristic of all types of SCID is absence of T-cell-mediated cellular immunity due to a defect in T-cell development.

    CD3 and active Caspase 3 populations 72 hrs after mouse aortic allograft

    C57Bl/6 donor aortic allografts were transplanted into Balb/C recipient mice (N = 3 per treatment) and followed up at 72hrs. Compared to saline, Serp-2 but not CrmA treatment reduced caspase 3 activity (panels A-C; p<0.0224). Neither protein treatment significantly reduced CD3+ T cells (panels D-F).

    This blot was produced using a 4-12% Bis-tris gel under the MES buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using Licor blocking buffer before being incubated with ab5690 and ab8245 (loading control) overnight at 4°C. Antibody binding was detected using Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) at a 1:10000 dilution for 1hr at room temperature and then imaged.

    IHC image of CD3 staining in a formalin fixed, paraffin embedded normal human tonsil tissue section*, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6) for 20 mins. The section was then incubated with ab5690 at 1/100 dilution for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    ab5690 staining CD3 in human lymph node tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded tissue sections). The sections were fixed in paraformaldehyde and subjected to heat-mediated antigen retrieval in citric buffer pH 6.0, prior to blocking with 10% serum for 1 hour at 20°C. The primary antibody was diluted 1/100 and incubated with the sample for 12 hours at 4°C. An HRP-conjugated goat anti-rabbit polyclonal was used as the secondary antibody, diluted 1/200.

Thank you for reply. Here the informations you need:

- Could you explain the probe conjugated secondary antibody? What color of the pobe should be?
> the conjugated probe was a florophore, Alexa-488, emitting in green. The image that i provided was in greyscale, showing the upper part of the brain cortex.
- Did you fixed the sections in acetone or PFA after putting these on slide?

>The whole mouse adult brain had been fixed in PFA 4% in PBS for 30minutes, then left in saccarose at 4°C. Later, it was cut at vibratome putting the slices in PBS. The immunostain staining protocol was performed with floatting sections. Sections had been mounted on the microscope glass only at final step.

- Please provide the purhcase order number and details about conditons of antibody storage?

Thank you for confirming the details.

This antibody has been positively used in many publications and in more than 25 positive Abreviews so I am sorry we are not sure the cause of the problem.

Also we have not tested this antibody with free floating Brain tissue sections so we are not aware about any non specificity. This antibody has been successfully used in free floating sections in the publication http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?cmd=Retrieve&db=PubMed&list_uids=20126467&dopt=Abstract. We ourselves do not hold any IHC-FoFr application data.

Interestingly in the following publication http://www.molbiolcell.org/content/19/6/2444.full.pdf Fig 2G the anti CD3 antibody seems staining the axonal processes. Please check.

Finally, I would like to suggest optimizing the protocol e.g. try changing the conc./dilution of primary antibody, try no primary control

I am sure with minor protocol tweaks the antibody would give you expected staining. If however the results do not improve, please do not hesitate to contact me for further assistance.

Inquiry: Dear Abcam, I tried antiCD3 Ab5690 on brain tissue. The antibody recongnizes axon processes in the cortex, that is not reported by litterature to me. I guess it is a aspecific signal. Protocol was following: -whole brain fixed in PFA, cut by vibratome. Tissue slices keept at -20°C in cryoprotecive solution. -washing in PBS -Blocking in fetal calf serum 10%, Triton X100 0.1% in PBS. -Antibody incubation overnight, at 4°C dilution 1/100 in blocking solution. -washes 3x20min, in PBS -Secondary antibody, probe conjugated dilution at 1/500 in blocking solution, 2hours at RT. -washes 3x20min, in PBS and mount.

Hello, I am using your anti-CD3 Ab and finding that I am getting staining of not only lymphocytes, but of smooth muscle cells as well. I previously tested the antibody on human tonsil sections, and it worked well. I used heat-mediated antigen retrieval in sodium citrate buffer, blocked with peroxide for 30 minutes, and then 10% normal goat serum for one hour. I used a 1:300 dilution of primary, incubated overnight at 4 degrees, washed thoroughly, and used a biotinylated goat anti-rabbit secondary at a concentration of 1:500 (incubated for 1 hour at 20 degrees). The sample was fixed using formalin and paraffin-embedded. Could you provide some feedback?

As stated on the datasheet, this antibody is not conjugated and has been used in combination with some HRP- conjugated secondary antibodies in our IHC experiments (see images).

As a Biotin-conjugated secondary I would suggest

Thank you for your response to my enquiry. I can confirm the purchase order number for the antibody was IR13304 and it was ordered on 9.7.12. While I was checking this I found that one of my colleagues had purchased the same antibody on the same day, so I tried an aliquot of hers and it worked beautifully! I can only presume that the antibody that I was sent was incorrectly labelled.
