    • Function

      Receptor for the Fc region of complexed or aggregated immunoglobulins gamma. Low affinity receptor. Involved in a variety of effector and regulatory functions such as phagocytosis of immune complexes and modulation of antibody production by B-cells. Binding to this receptor results in down-modulation of previous state of cell activation triggered via antigen receptors on B-cells (BCR), T-cells (TCR) or via another Fc receptor. Isoform IIB1 fails to mediate endocytosis or phagocytosis. Isoform IIB2 does not trigger phagocytosis.
    • Tissue specificity

      Is the most broadly distributed Fc-gamma-receptor. Expressed in monocyte, neutrophils, macrophages, basophils, eosinophils, Langerhans cells, B-cells, platelets cells and placenta (endothelial cells). Not detected in natural killer cells.
    • Involvement in disease

      Note=A chromosomal aberration involving FCGR2B is found in a follicular lymphoma. Translocation t(1;22)(q22;q11). The translocation leads to the hyperexpression of the receptor. This may play a role in the tumor progression.
    • Sequence similarities

      Contains 2 Ig-like C2-type (immunoglobulin-like) domains.
    • Domain

      Contains 1 copy of a cytoplasmic motif that is referred to as the immunoreceptor tyrosine-based inhibitor motif (ITIM). This motif is involved in modulation of cellular responses. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases.

      Ich kann Ihnen nun bestätigen, dass beide Antikörper spezifisch die IIb Isoform (und alle Unterformen von dieser Isoform: IIb1, IIb2, IIb3) binden.

      Beide Immunogen sind Peptide die die folgende Sequenz einschließt: TITYpSLL.

      Die Antikörper wurden nicht experimentell mit Isoform IIa und IIc getestet. Aber diese beiden Isoformen enthalten die Immunogensequenz TITYpSLL nicht.

      ab68423 detects CD32b and is not expected to cross-react with CD32a. However, this has not been specifically tested in the laboratory as far as I am aware.

      Inquiry: I noticed that the Epitomics data sheet for this antibody (clone EP926Y) states that it cross reacts with mouse CD32b (phospho Y292), as determined by western blot. Abcam states that it does not cross react with mouse or rat. I'm wondering which is true, or why there is this descrepancy. This is in reference to Abcam # ab68423 or Epitomics # 2308-1. Thanks.

      Thank you very much for contacting us with your question.

      I apologize for the confusing discrepancy on the Abcam and Epitomics datasheets. I've gotten in touch with the Epitomics lab, and they did test the antibody once on mouse samples (brain, heart, kidney and spleen) at a 1:100,000 primary dilution but did not observe a signal. They still recommend the product for mouse reactivity based on the fact that the immunogen sequence has 100% homology with the mouse protein.

      Though Epitomics is now an Abcam company, we are still working to merge all of our policies. Abcam only lists a species as reactive if it has actually been tested and shown positive results. If the results were negative then we say the antibody does not react with that species.

      Due to the above information, I hesitate to recommend this antibody for use with mouse samples. There may be tweaks to the protocol that would produce a signal with mouse lysates (such as increasing the concentration, 1:100,000 is quite high) but we can not guarantee this antibody to work with mouse.
