FunctionPutative adhesion molecule of myelomonocytic-derived cells that mediates sialic-acid dependent binding to cells. Preferentially binds to alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. In the immune response, may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signaling molecules. Induces apoptosis in acute myeloid leukemia (in vitro).

CD33 was immunoprecipitated from 0.35 mg THP-1 (Human monocytic leukemia monocyte) whole cell lysate 10ug with ab264558 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab264558 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366) was used at 1/5000 dilution.
Lane 1: THP-1 (Human monocytic leukemia monocyte) whole cell lysate 10ug
Lane 2: ab264558 IP in THP-1 whole cell lysate
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab264558 in THP-1 whole cell lysate
Blocking and dilution buffer and concentration: 5% NFDM/TBST.

Flow cytometric analysis of Human peripheral blood mononuclear cell (PBMC) cells labelling CD33 with ab264558 at 1/50 (Right) compared with a Rabbit monoclonal IgG (ab172730) isotype control (Left).
Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) at 1/2000 dilution was used as the secondary antibody.
Cells were stained with rabbit IgG (Left) or ab264558 (Right). Then stained with anti-CD14 conjugated to Alexa Fluor® 647 (PMID: 26468886).
Gated on viable cells.
Flow cytometric analysis of Jurkat (Human T cell leukemia T lymphocyte, Left) / THP-1 (Human monocytic leukemia monocyte, Right) cells labelling CD33 with ab264558 at 1/50 compared with a Rabbit monoclonal IgG (ab172730) / Black isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue).
Goat anti rabbit IgG (Alexa Fluor®488, ab150077) at 1/2000 dilution was used as the secondary antibody.
Gated on viable cells.
