  • Function

    Possible adhesion molecule with a role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. Could act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. Presents carbohydrate ligands to selectins.

      IHC image of CD34 staining in Human normal placenta formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab762, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    Ich habedas Labor kontaktiert, jedoch ist diese Kontrolle leider nicht kommerziell erhältlich. Diese transfektierten Zellen sind vom Labor die diesen Antikörper produziert, und sind leider nicht zum Verkauf erhältlich.

    Ich kann Ihnen jedoch auch Pankreas-, Nieren-, Hirn- oder Plazenta-lysatals positive Kontrolle empfehlen, da diese Organe ein gutesExpressionsniveau von CD34 haben.

    Vielen Dank für Ihre Email und die Protokollinformationen. Es tut mir leid, dass Sie Probleme mit unserem Antikörper hatten.

    Ich habe mirdas Protokoll angeschaut, und mit unseren Daten verglichen. Umdas Problem zu lösen, möchte ich die folgenden Vorschläge zu Veränderung des Protokolls machen:

    1.) Wir empfehlen diesen Antikörper unter nicht reduzierten Konditionen zu benutzen. In der Tat scheint ansonsten das Epitope verloren zu gehen. Ich kann daher empfehlen die Proben wie folgt zuzubereiten:
    ResuspendierenSie circa50MillionenZellencells in 1 mlkaltem Lysis Puffer (1% laurylmaltoside in 20 mM Tris/Cl, 100 mM NaCl pH 8.2, 50 mM NaF mit Protease Inhibitor Cocktail). InkubierenSie danndies60 Minuten auf Eis.Zentrifugieren umZelldebris zu entfernen.Fügen Sie dann die entsprechende Menge des nicht-reduzierendenLoading Puffers hinzu um die Proben auf das SDS PAGE zu laden.

    2.) Obschon ich denke, dass die andere Probenzubereitung das Problem lösen wird, kann ich auch empfehlen, alternative Blockierungspuffer zu benutzen. In der Tat kann das Ändern des Blockierungspuffer das Resultat stark beeinflussen. Ein Beispiel dafür ist auch das Bild auf dem Datenblattdes ab9385.https://www.abcam.com/HRP-GAPDH-antibody-Loading-Control-ab9385.html (or use the following: https://www.abcam.com/HRP-GAPDH-antibody-Loading-Control-ab9385.html). Ich kann Ihnen daher empfehlen, anstelle der Milch auch BSA auszuprobieren.

    wir haben Ihren oben genannten Antikörper für Western Blot –Analysen von KG1a Zellen verwendet und konnten auch nach einigen Variationen in der Menge des eingesetzten Proteins sowie bei der Verdünnung des AK leider keine positiven Ergebnisse erhalten.

    Nun wollte ich bei Ihnen nachfragen, wie viel Protein Sie bei Ihren Analysen eingesetzt haben, um positive Ergebnisse zu erzielen bzw. ob Sie noch einen anderen Tipp für uns haben?!

    Es tut mir leid zu hören, dass Sie Probleme mit diesem Antikörper haben. Normalerweise, sollten 10ug Lysat genügend sein, um eine Bande zu detektieren. Dies hängt natürlich jedoch vom Lysat und dem Protokoll ab. Auf dem Datenblatt von ab63985 hat es ein schönes Bild eines Westernblots ggen CD34. https://www.abcam.com/CD34-antibody-ab63985.html

    Ich würde gerne helfen, das Problem zu lösen. Ich habe Ihnen deshalb unseren Fragebogen als Word-Dokument an diese E-Mail angehängt. The material safety datasheets are issued based on the inclusion of additional components such as Na Azide and Glycerol in addition to specific componds that we sell for particular techniques such as DAB for IHC etc. I hope this makes sense.
