    Mouse monoclonal [QBEND-10] to CD34

    This antibody recognizes the CD34 antigen, some cross reactions with vascular associated adventitia and some basement membranes. Ultrastructure studies, using an indirect immuno-gold technique on ultra thin sections of human umbilical artery, have revealed that this antigen is associated to a degree with the basoluminal surfaces of endothelium and shows a slight reaction with an intracytoplasmic vesicular compartment. Predominant staining is on the endothelial cell membrane. QBEND-10 immunoprecipitates a glycoprotein with a relative MW 110kD expressed on haematopoietic cells and on the established myeloid leukaemic cell line KG1A.

    Possible adhesion molecule with a role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. Could act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. Presents carbohydrate ligands to selectins.

      Overlay histogram showing Jurkat cells stained with ab8536 (red line). The cells were fixed with methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab8536, 1/50 dilution) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a significantly decreased signal in Jurkat cells fixed with 4% paraformaldehyde (10 min) used under the same conditions.

      ab8536 staining CD34 in human kidney tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 2% BSA for 10 minutes at 21°C; antigen retrieval was by heat mediation in a citrate buffer. Samples were incubated with primary antibody (1/2000 in blocking buffer) for 16 hours at 21°C. A Biotin-conjugated goat anti-mouse IgG polyclonal (1/300) was used as the secondary antibody.

    Here is the additional information for flow cytometry - in addition to the figure legend given on the datasheet:

    Methanol is used at 80% to fix the cells.The cells are blocked for 30minutes and there is a washing step between addition of the primary and secondary.

    The Exact epiotpe site for ab8536 has not been mapped, so we are unsure if the epitope is intra- or extracellular. However we do know that the predominant staining is on the endothelial cell membrane.

    We have not tested ab8536 in flow cytometry in non-fixed cells, it may work in non-fixed cells but as we have not tested it, we cannot guarantee that it will work for you.

Can we suggest alternatives as ab34364, ab87951 and ab101500? ab33775; IHC-P is not a tested application we are unable to confirm its use. We suggest using alternative as ab846, ab51312 ab33854; IHC-P again is not a tested application for this ab. We can suggest using ab9496, ab59754 In case you are interested in buying ab65865, ab33775 and ab33854 only I can then offer you three Abreview discount codes which will give you next three antibodies free of charge. Please be advised that in some cases heat damages the antigenic epitope we therefore always recommend our customers optimizing the heat mediated antigen retrieval step.     Thank you for your enquiry. This antibody (ab8536) is not purified. However, it is at a higher concentration now.
