      Question

      We have purchased some antibodies against rat platelets:

      Primary:

      ab108949 - mouse monoclonal anti CD63, FITC labeled
      (ab106163 as isotype control)

      ab54427 - mouse monoclonal anti CD62P
      (ab91353 as the isotype control, as a secondary antibody)

      ab95780 anti CD42d armenian hamster antibody
      (ab18479 as an isotype control, for which we are using APC-conjugated Goat Anti-Hamster IgG from Leinco).

      We are ibodies.doing flow cytometry experiments on fixed whole blood, and fixed platelet-rich plasma. The problem is that the samples with the isotype control show the same fluorescence as the samples with the primary ant We have tried a number of different fixing and staining protocols, with and without washing, but until now had no success. What I am wondering about is, how reliable are these primary antibodies for the use with rat platelets?

      I am sorry that you have been having issues with some of our antibodies in Flow Cytometry. Under our Abpromise we guarantee that ab108949, ab54427 & ab95780 should work in flow cytometry on rat samples. If we cannot resolve the issue you are having with the antibody then I would be happy to either send a replacement antibody or to process a refund.

      To be able to help you further in this matter, would you be able to send me a copy of the protocol that you have been using, along with any modifications that you have tried, to see if there is anything I can suggest.

      For the secondary antibody, will the first one (ab6947) work against ab54427, or do we need to buy the second one (ab97239)? One is against IgG, the other one is against IgG1. ab54427 is IgG1.
