      ab25340 can inhibit hyaluronate-dependent cell aggregation, prevent lympho-hemopoiesis in both Dexter and WhitlockWitte Cultures, prevent the earliest intrathymic precursors from homing to the thymus and costimulate the activation of freshly purified splenic CD4+ T cells.

    Target

    • Function

      Receptor for hyaluronic acid (HA). Mediates cell-cell and cell-matrix interactions through its affinity for HA, and possibly also through its affinity for other ligands such as osteopontin, collagens, and matrix metalloproteinases (MMPs). Adhesion with HA plays an important role in cell migration, tumor growth and progression. Also involved in lymphocyte activation, recirculation and homing, and in hematopoiesis. Altered expression or dysfunction causes numerous pathogenic phenotypes. Great protein heterogeneity due to numerous alternative splicing and post-translational modification events.
    • Tissue specificity

      Isoform 10 (epithelial isoform) is expressed by cells of epithelium and highly expressed by carcinomas. Expression is repressed in neuroblastoma cells.
    • Sequence similarities

      Contains 1 Link domain.
    • Domain

      The lectin-like LINK domain is responsible for hyaluronan binding.
    • Post-translational
      modifications

      Proteolytically cleaved in the extracellular matrix by specific proteinases (possibly MMPs) in several cell lines and tumors.
      N-glycosylated.
      O-glycosylated; contains more-or-less-sulfated chondroitin sulfate glycans, whose number may affect the accessibility of specific proteinases to their cleavage site(s).
      Phosphorylated; activation of PKC results in the dephosphorylation of Ser-706 (constitutive phosphorylation site), and the phosphorylation of Ser-672.

      I am sorry I have over-read your main question. We state only mouse as a tested species on the datasheet so far. All tested applicationsare specified on our datasheets, and these are updated as soon as any new information is brought to our attention.Therefore, it is unlikely that this antibody has been tested on human tissue so far.

      However, I performed a BLAST search in which I compared the mouse CD44 Isoform 1 with the human protein and most of the human isoforms have a very high homology ( between 87 and 100%), therefore, I am positive that the antibody should detect in human tissue. Unfortunately, we cannot guarantee that it will work.

      For the future, if you would like to test the antibody inan untested application or species, please contact our Scientific Support team prior to the purchase as you may be eligible for our testing discount program.
