    Target

    • Function

      Protein tyrosine-protein phosphatase required for T-cell activation through the antigen receptor. Acts as a positive regulator of T-cell coactivation upon binding to DPP4. The first PTPase domain has enzymatic activity, while the second one seems to affect the substrate specificity of the first one. Upon T-cell activation, recruits and dephosphorylates SKAP1 and FYN.
    • Involvement in disease

      Defects in PTPRC are a cause of severe combined immunodeficiency autosomal recessive T-cell-negative/B-cell-positive/NK-cell-positive (T(-)B(+)NK(+) SCID) [MIM:608971]. A form of severe combined immunodeficiency (SCID), a genetically and clinically heterogeneous group of rare congenital disorders characterized by impairment of both humoral and cell-mediated immunity, leukopenia, and low or absent antibody levels. Patients present in infancy recurrent, persistent infections by opportunistic organisms. The common characteristic of all types of SCID is absence of T-cell-mediated cellular immunity due to a defect in T-cell development.
      Genetic variations in PTPRC are involved in multiple sclerosis susceptibility (MS) [MIM:126200]. MS is a neurodegenerative disorder characterized by the gradual accumulation of focal plaques of demyelination particularly in the periventricular areas of the brain. Peripheral nerves are not affected. Onset usually in third or fourth decade with intermittent progression over an extended period. The cause is still uncertain.

        Lewis rat splenocytes stained with ab6329 (right) or mouse IgG2aκ (left). Lewis rat splenocytes were incubated for 30 min on ice in 10% rat serum to block FC receptors and non-specific protein-protein interaction followed by the antibody (ab6329) or mouse IgG2aκ Isotype (ab18450) (1x106 in 100µl at 0.2 µg/ml) for 30 min on ice.

        The secondary antibody Goat Anti-Mouse IgG H&L (Alexa Fluor ® 488, pre-adsorbed) (ab150117) was used at 1/2000 dilution for 30 min at 4°C. The cells were simultaneously stained with CD3 APC antibody.

        Acquisition of >30,000 events were collected using a 50 mW Blue laser (488nm) and 530/30 bandpass filter. Events were gated on viable lymphocytes.

      Thank you for your enquiry. Whilst we do not have a recommended positive control the following publication employed leukocytes as a control for the detection of CD45 transcripts in rat cells. De Dios I, Ramudo L, Alonso JR, Recio JS, Garcia-Montero AC, Manso MA. CD45 expression on rat acinar cells: involvement in pro-inflammatory cytokine production.       You recently spoke with my colleague Maud Osborne regarding the use of a number of anti CD antibodies in IHC on paraffin embedded rat sections. As you have seen, none of the antibodies had been tested in the exact conditions you wish to try although my comments about their use are below. ab26239 (CD44) - has been tested in IHC-P on mouse & human although not yet tested in rat. As it has been shown to work in mouse I think it's a good bet that it will work in rat although there's no guarantee. ab6329 (CD45) - has been tested in IHC on Frozen sections but not on paraffin sections and shown to cross react with rat. ab22479 (CD45) - raised in rat and shown to cross react with rat - take care when using this antibody in IHC to block any endogenous rat IgGs that may bind to your anti rat secondary ab23844 (CD44) - works in IHC-P and cross reacts with cow, dog, goat, horse, pig, sheep so again although there's no guarantee, I think it's reasonable to think that this will cross react with rat too! ab19673 (CD105) - cross reacts with human and works in IHC-P I hope this is a helpful summary.

      Thank you for your patience. This product has been tested in IHC-frozen, used at dilutions between 1:25 - 1:200. I have updated the datasheet to reflect this information for future use. To the best of my knowledge, it has not been tested in IHC-paraffin. I hope this information helps, please do not hesitate to contact us if you need any more advice or information.

      This antibody is raised in mouse and so is difficult to use on mouse tissue without getting problems with background staining. We have no records of this antibody working on paraffin sections.
