      ICC image of ab59479 stained A431 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody ab59479 at 5µg/ml overnight at +4°C. The secondary antibody (green) was DyLight® 488 goat anti- mouse (ab96879) IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

      IHC image of CD63 staining in Human Skin Melanoma formalin fixed paraffin embedded tissue section, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab59479, 5 µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

       

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

      Overlay histogram showing HL60 cells stained with ab59479 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab59479, 1 µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight®488 goat anti-Mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was Mouse IgG1 [ICIGG1] (ab91353, 2 µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in HL60 cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

    Question

    so for ab8219 you are not aware of any robust and reproducible Western data, but with ab59479 based on your experience its guaranteed that Western will work fine. You think depending on the cell line (and we'll also use exosomes from blood) we'll see several bands in the range 30-60 KDa?  We purchased both antibodies from abcam already, but Western conditions are not optimized yet. With 8219 we see one band ~ 42 KDa, another > 180 kDa. For ab59479 we see band ~40 KDa, just one faint band. Both gels were denaturing, non-reducing conditions. thanks,

    In regards to ab8219, I have had the chance to talk with the lab. According to them this antigen is indeed very variable on WB of various material. I am sending the pattern they obtained on non-reducing K562 cells (see image CCE3541736_MEM-259WB.jpg), however observing more heavier bands using other material would be also a reasonable result.

    I have attached information given to me by the lab producing ab59479 (see remainng attached). These include a puplication which has used the product in Western blot as well as a Powerpoint file which contains lab testing results.

    Dear Sir/Madam: We need an anti-human CD63 antibody, to be used for Westerns. Specifically, we plan to detect CD63 protein from the human exosomes isolated from blood and cell culture media. Which antibody do you recommend- ab8219, ab59479 or other? PI sheet for Ab8219 anti-CD63 says that specificity is CD63, 25-26 kDa (or larger if glycosilated); But PI sheet for Ab59479 that is also anti-CD63 says that specificity is LIMP antigen, a 30-60 kDa (smear) protein; Thank you very much!

    Answer

    Thank you for contacting us.

    CD63 antigen is also known as LIMP and LAMP-3. While unmodified protein is about 25kDA, this is heavily glycolylated. AB59476migrates like a smear due to a very complexe N-glycosylation. I would recommend this product because this real world information is available whereas the data for ab8219 appears to be based on theory and not practise.

    Please remember that differences exist between the cell lines (e.g. K562 givesbetter migration than HEL).

    It is important to remember that this antibody (as well that all theanti-tetraspanine antibodies) must be used under non-reduced conditions.
