    CD77 is specific for Burkitt's lymphoma associated antigen (BLA), which is a neutral glycosphingolipid of the globoseries: globotriaosylceramide Gb3. Reacts with red blood cells of phenotype pk1 or pk2 and activated B cells in agglutination assay at 4°C and the radioactive Coombs test. Does not react with P or P2 red cells, slightly agglutinate P1 red cells after papain treatment (anti pk activity).

  • Overlay histogram showing RAMOS cells stained with ab19795 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then incubated in 1X PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab19795, 1/10 dilution) for 30 min at 22ºC. The secondary antibody used was Goat Anti-Rat IgM mu chain (DyLight® 488) preadsorbed at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was rat kappa monoclonal [RTK2118] (ab35768, 2µg/1x106 cells ) used under the same conditions. Acquisition of >5,000 events was performed.

The laboratory that supplies this to us does not check the concentration of the antibody solution, so matching an isotype control concentration to the concentration of the antibody will not be possible, unless you are able to measure the concentration yourselves, for instance by BCA or an absorbance measurement.

We do not have an optimal dilution recommendation for IHC. The 1/100 dilution I mentioned may be a good place to start but the best approach is to test a dilution series to find which dilution gives the best specific signal to background ratio, for instance, 1/25, 1/75, 1/225.
