Involvement in diseaseDefects in CD8A are a cause of familial CD8 deficiency (CD8 deficiency) [MIM:608957]. Familial CD8 deficiency is a novel autosomal recessive immunologic defect characterized by absence of CD8+ cells, leading to recurrent bacterial infections.

FormCD8 beta tissue specificity: Isoform 1, isoform 3, isoform 5, isoform 6, isoform 7 and isoform 8 are expressed in both thymus and peripheral CD8+ T-cells. Expression of isoform 1 is higher in thymus CD8+ T-cells than in peripheral CD8+ T-cells. Expression of isoform 6 is higher in peripheral CD8+ T-cells than in thymus CD8+ T-cells. CD8 beta PTM: Phosphorylated as a consequence of T-cell activation.

ab4055 staining CD8 in human spleen tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 2% BSA for 10 minutes at 21°C; antigen retrieval was by heat mediation in citric acid. Samples were incubated with the primary antibody (1/100 in TBS/BSA/azide) for 16 hours at 21°C. A biotin-conjugated goat anti-rabbit IgG polyclonal (1/300) was used as the secondary antibody.
ab4055 staining CD8 in Human colon tumour tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with paraformaldehyde, permeabilized with 0.3% Triton X-100 in PBS and blocked with 5% serum for 1 hour at 24°C. Samples were incubated with primary antibody (1/500 in PBS + 0.3% Triton X-100 + 1% BSA) for 3 hours at 24°C. An undiluted Alexa Fluor® 488-conjugated Goat anti-rabbit IgG polyclonal was used as the secondary antibody. Left - patient with low CD8, Right - patient with high CD8.
