      IHC image of ab47153 staining in human tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab47153, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    I am sorry to confirm that the epitope for these antibodies have not been mapped. This is not often done for research grade antibodies.

    I can confirm that the immunisation was done with transfected cells. So the immunogen was the full sequence, expressed at the membrane.

    Thank you very much for your reply.

    I understand that you dont guarantee the tests that you dont cover in your studies. But it is strange that the antibody didn’t work in my experiments only with the last two vials. Could I please ask if I place an order for the same antibody if it is possible to get a vial from either the same old aliquot that I had before the last two ones, namely GR45737-1, Or if I can at all get a totally new lot number??

    We currently only have GR45737-2 and GR45737-3 in stock. We have sold out of GR45737-1. However, these are all the same antibody and are all from the same parent lot, just aliquoted at different times. If you would like to order a specific lot, you'll have to mention that to customer service when ordering. Lot GR45737-2 would ship from our US office and GR45737-3 would ship from our UK office.

    Question

    I have been using anti Kai1 antibody (Anti-CD82 antibody [B-L2] - Azide free (ab47153)) for quite some time. I use it to block the interaction of the protein with another protein in cell culture.

    The antibody was working fine until I received the last two vials. They both have the same lot number: GR45737-3. I ordered the second vial cause the first one didn’t work but now I found out that the second one doesn’t work either.

    Would you please be able to help me to either find out whether there is something wrong with this batch or perhaps you could send me another vial that I can try? I have been ordering quite a lot of this particular antibody and never had a problem until only recently and because I need it quite urgently your prompt response is highly appreciated.

    Unfortunately, we have not tested, nor do we guarantee, this antibody to work in blocking protein-protein interactions. If we had, we would list that on the online datasheet as "blocking," "functional studies," or "neutralizing." In this case, this antibody has only been tested in IHC-P, IHC-Fr, ICC/IF, andFlow Cyt.


    Since the antibody is being used in an untested application, we cannot offer a replacement or refund in accordance with our Abpromise.

    It looks like none of our anti-CD82 antibodies have been tested for their blocking ability and the only lots we have in stock for ab47153 are from the same parent lot as GR45737-3.


    I see that you have ordered this antibody 4 times in the past. You've tried lot GR45737-1and GR45737-3. These are from the same parent lot. The number after the dash just describes the order in which it was aliquoted. I do not know why, then, that you would be seeing different results as these are in fact from the same parent lot of antibody.
