    Thank you for your patience. The lab has confirmed that ab92574 was tested for Flow Cytometry and gave negative results. We do not have any antibodies for CD90 that are known to give good signal in rat but no signal in mouse, and unfortunately I do not know of any markers the customer could use to distinguish the mouse cells from the rat cells in their assay. I am sorry I could not be more helpful, please let me know if you have any further questions.

    Thank you for submitting the completed questionnaire and for including the ICC images.

    It seems like the few cells that are stained is just the result of non specific background staining.

    A couple additional questions:

    1. How long was the primary antibody incubated with the sample, and at what temperature?

    2. Were other antibodies used successfully to stain the same cells?

    3. Is there evidence that CD90 is expressed in this cell type?

    I apologize for the confusion of the datasheet for ab92574. After checking our notes about the non-reactivity with mouse, I found that this antibody does work on mouse samples in WB, but has not worked in mouse samples in other applications. The rat cross-reactivity has only been validated in WB. I have updated our datasheet to reflect this information and have removed mouse as a non-reactive species.

    I was unable to find any notes about why this antibody failed in Flow Cytometry. I have emailed the lab to see if they have any additional information about this testing and I will let you know as soon as I hear from them.

    Thank you for your email. The overall sequence homology for the full length proteins are: human-cow: 74% mouse-cow: 69% rat-cow: 70% The majority of our CD90 antibodies were raised against full length proteins or cell lines and therefore the epitopes are not known. Only ab92574 and ab110477 were raised agianst peptides. For ab92574, we know the homology is poor. For ab110477 I have inquired with the lab as I currently have no detailed information about the immunogen sequence. I will let you know what I find out. Here is the link to the datasheet for ab110477: https://www.abcam.com/CD90-Thy1-antibody-ab110477.html It has so far only been tested in WB with human samples. Should the homology be high for the immunogen, I'd be happy to send you a discount code for trying the antibody in IHC-P, bovine and sheep. I hope this information is somewhat of help so far.
