ChIP Use at an assay dependent concentration.
ICC/IF Use a concentration of 4 µg/ml. Fix with 4% paraformaldehyde.
IP Use at an assay dependent concentration.
WB Use a concentration of 1 µg/ml. Detects a band of approximately 38 kDa (predicted molecular weight: 35,38,20 kDa).

Target

  • Function
    Important transcriptional activator in the regulation of genes involved in immune and inflammatory responses. Specifically binds to an IL-1 response element in the IL-6 gene. NF-IL6 also binds to regulatory regions of several acute-phase and cytokines genes. It probably plays a role in the regulation of acute-phase reaction, inflammation and hemopoiesis. The consensus recognition site is 5'-T[TG]NNGNAA[TG]-3'. Functions in brown adipose tissue (BAT) differentiation.

We have not tested ab15050 in ChIP, therefore we cannot guarantee that it will work in this application. Since it does work for IP though, there is a good chance that it will be successful.

Thank you for getting back to me on this.

I have put some red arrows next to what I believe are the correct size bands. It looks to me as though the correct bands are present in the mouse cell lysate (although faint) and that in your samples the 38 & 35kDa bands are present. It’s hard to say if the 20kDa band in just not present in your samples, or it present but a low levels. Is it possible that your samples would not contain the 20kDa form?

To try and clean up the blot a little, I would recommend, unless you are already doing so in to use a 5% BSA block and also incubate your primary antibodies in the BSA as well. Also, increase the percentage of tween in your wash buffer to 0.1% and see if that helps to clean up the membrane.

I have looked at the image you sent me and you are getting the correct banding pattern, but everything is 10-15kDa higher than it should be. So you could be really unlucky and the antibody is binding non-specifically (but still giving the triplicate banding), which I doubt, or the bands you are seeing are the correct bands, but for whatever reason are at a higher molecular weight.

I think the best way to try and determine what is going on is for me to send you ab14632, 3T3-L1 (Mouse embryonic fibroblast - adipose like cell line) Nuclear Lysate.

To send you the cell lysate, could you please either me your full shipping address, or send me the original order number for ab15050, so that I can get the address from there.
