      Cell Tracking Dye Kit - Red | Cytopainter (ab138893) is designed to uniformly label live cells for studies where multicolor analysis might be required, such as cell drug resistance, cell adhesion or chemotaxis. Dye can be visualized with a blue filter (Ex/Em = 575/600 nm).


      The labelling process is robust, requiring minimal hands-on time. This product can be used in different types of fluorescence platforms such as microplate assays, flow cytometry and fluorescence microscope.The kit provides all the essential components with an optimized cell-labelling protocol, and can be used for both proliferating and non-proliferating cells (either suspension or adherent cells).

    • Notes

      Uniform labeling of live cells with a hydrophobic dye whose fluorescence is strongly enhanced upon entering the cell. Cellular esterases convert the substrate into a strong fluorescent photostable hydrophilic dye that is well retained in the cytoplasm. Cells need to be viewed in less than 24 - 48 hours as dye will be passed to daughter cells.

    A great cell labeling reagent

    This product worked straight as written on the website in the sea urchin eggs and embryos. 10-minute incubation was good enough to label eggs (attached image) and the fluorescence lasted at least for 36 hours. No complex preparation was needed.

    Question

    I have a problem with it because I observed a lot of red discrete points and not a diffuse staining of the cytoplasm. I think the staining is blocked in endosomes.
    I would like to know if you already had feedback with this problem on primary cells? What can be the explanation?

    I have contacted the lab and they have confirmed that this staining pattern observed is to be expected because the dyes used in cell tracking kits can accumulate in any cell organelles such as cytoplasm (preferably), mitochondria, and lysosome etc. In this particular case, the dye has mostly accumulated in the lysosome. Other kits from other suppliers will differ in that the dye will accumulate in different organelles.

    We are currently trying to develop a kit which uses a dye which accumulates predominantly in the cytosol which might be more useful to you. Alternatively, to see a more diffuse staining all over the cells as seen on our datasheets, a high concentration of the dye will need to be used.
