Target

  • Function

    Involved in the transfer of insoluble cholesteryl esters in the reverse transport of cholesterol.
  • Tissue specificity

    Expressed by the liver and secreted in plasma.
  • Involvement in disease

    Defects in CETP are a cause of hyperalphalipoproteinemia (HYPALIP) [MIM:143470]. Affected individuals show high levels of alpha-lipoprotein (high density lipoprotein/HDL).
    Defects in CETP are the cause of cholesteryl ester transfer protein deficiency (CETP deficiency) [MIM:607322]. This is an autosomal dominant condition associated with increased HDL cholesterol levels.

      Immunocytochemistry/Immunofluorescence analysis of CETP in HepG2 cells. Cells were grown on chamber slides and fixed with formaldehyde prior to staining. Cells were probed without (control) or with ab2726 (1:20) overnight at 4°C, washed with PBS and incubated with a DyLight-488 conjugated secondary antibody. CETP staining (green), F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue) is shown. Images were taken at 60X magnification.

    Question

    One customer have bought one vial ab2726(CETP antibody ), the lot No is 72205. She used it to test CETP expressed in COS7 cells( transfected with full lenght human or animal CETP gene) at 1:400(recommendated by datasheet). But the result was not good: no bands with short exposure time, then elonged to 15mins, several bands appeared with high backgroud. the detect system is proved to work with other primary antibody. Then, she used another CETP polyclonal antibody prepared by themself to test as contrast. the result is good, clear target bands can be showed( see attchment for result picture) . Can you give me your suggestions to solve this problem asap?
