  • ab15160 staining Chromogranin A in mouse gastric gland cells by Immunocytochemistry/ Immunofluorescence. Cells were fixed with paraformaldehyde, permeabilized with 0.2% Triton ×100 and blocking with 10% serum for 45min at 200C was performed. Samples were incubated with primary antibody (1/100) for 18 hours at 4°C. A Texas Red ®-conjugated goat polyclonal to rabbit IgG was used at dilution 1/200 as secondary antibody.

  • ab15160 (1/100) staining Chromogranin A in paraffin-embbeded mouse pancreas tissue sections. Tissue underwent fixation in formaldehyde, heat-mediated antigen retrieval in citrate buffer pH 6.0 and blocking (10 minutes, 1.5% serum). An HRP-conjugated goat polyclonal was used as the secondary. For further experimental details please refer to abreview.

  • ab15160 staining Chromogranin A in mouse colon tissue section by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue underwent formalin fixation before heat mediated antigen retrieval with 10mM citric acid buffer and then the sample was blocked for 45 minutes at 20°C. The primary antibody (1/100) was incubated with sample in 10% serum for 18 hours at 4°C. A Biotin conjugated goat polyclonal to rabbit IgG was used at dilution 1/200 as secondary antibody.

Answer

We recommend ab25693. This is currently the only chromogranin antibody for which we have canine IHC data. A collaborator tested it on sections of formalin-fixed paraffin-embedded canine small intestine and saw strong positive staining of a sub-population of cells. Here is a link to the review.

Answer

Thank you for your inquiry.

The immunogen sequence for ab15160 proprietary information, but I am happy to confirm it is located within aa 220-457 of P10645 (Uniprot). The epitope has not been mapped, thus the specific binding site is unknown.

We do not have any experimental data regarding the cross-reactivity of ab15160 with vasostatin, pancreastatin, serpinin, or catestatin. By sequence homology ab15160 should not recognize vasostatin. There is 100% homology between the immunogen for ab15160 and pancreastatin, serpinin, and catestatin so theoretically E1524 may recognize all of them.

Please do not hesitate to contact me with additional questions or concerns.

Answer

Thank you for your enquiry.

I am sorry to confirm we do not have any experimental data regarding the cross-reactivity ofab15160 with serpinin, parastatin or catastatin. There is 100% homology between the immunogen and all three cleavage products so theoretically the antibodymay cross-react with them, but this has not been tested in the laboratory.

If you still have some concerns regarding your results, please do not hesitate to contact me with further information, including details ofwhich application youare using. Our products are covered by a 6 month guarantee in the tested applications and species listed on the individual datasheets. We will be pleased to investigate this further for you.

I hope this will be helpful. Please do not hesitate to contact me with anyadditional questions.
